Effects Of Broccoli Superfine Powder On Growth Performance And Immune And Antioxidant Functions Of Broilers

Objectives: To study the effects of Calyx Seu Fructus Physalis ultramicro-pulverised powder on growth performance,immune function,oxidation resistance and intestinal health of broilers,comprehensively evaluate its application on broiler breeding as well as to develop it as a natural plant feed additives.Methods: Based on the principles,200 AA broilers of 7 days old with body weight 90-100 g were selected and randomly divided into 5 groups(each with 4 replicates and 10 chickens per replica).The blank control group was only fed with a basic diet while the other groups along with a basic diet were fed 0.005%chlortetracycline(antibiotic group),0.16% Calyx Seu Fructus Physalis ultramicro-pulverised powder(experimental group I),0.32% Calyx Seu Fructus Physalis ultramicro-pulverised powder(experimental group II)and 0.48% Calyx Seu Fructus Physalis ultramicro-pulverised powder(experimental group III)for14 days.After 14 days,the relevant indicators of production performance were obtained through the statistical weight and feed consumption data;the immunoglobulin and antioxidant indicators in the serum of the broiler chicken were measured by the kit;the intestinal tracts were observed by HE staining,a typical microscopic field was photographed to measure the height(V)and crypt depth(C)corresponding to each villus,and the intestinal villus height/crypt depth(V/C)was calculated;q RT-PCR was using to detect m RNA relative expression of intestinal tight junction proteins Claudin-1,Occludin,and ZO-1;bacterial16Sr DNA high-throughput sequencing of cecum contents of broilers were detected using the Miseq PE300 platform.Results: 1.Compared with the control group,the effect of Calyx Seu Fructus Physalis ultramicro-pulverised powder on the growth performance showed that,the average daily gain(ADG)and daily intake(ADFI)of antibiotic group and test II group was significantly higher(P < 0.05)but the difference between the two groups was not significant(P > 0.05).The F/g of antibiotic group,test group I and test group II were decreased significantly(P < 0.05).The result shows Calyx Seu Fructus Physalis ultramicro-pulverised powder increase the average daily gain of broilers and reduce the F/C ratio,thereby improving the growth performance of broilers.2.Compared with the control group,the effect of Calyx Seu Fructus Physalis ultramicro-pulverised powder showed that addition of Calyx Seu Fructus Physalis ultramicro-pulverised powder to the diet significantly improved the bursal index,spleen index and the contents of Ig A,Ig G and Ig M in the serum(p<0.05);The activity of GSH-Px and SOD in the antibiotic group and test II group were significantly increased(p<0.05)while T-AOC was significantly higher in all the four groups compared with the control group(p<0.05).The results showed that the Calyx Seu Fructus Physalis ultramicro-pulverised powder not only increases broiler's bursa index,spleen index,and serum Ig A,Ig G,Ig M content,enhances the immune function;it also improves broiler serum GSH-PX,SOD activity and T-AOC,enhances the oxidation resistance.3.Compared with the control group,the addition of Calyx Seu Fructus Physalis ultramicro-pulverised powder(0.32%)significantly increased the villi height and V/C of jejunum and ileum,and reduced the crypt depth of jejunum.Secondly,Occludin m RNA relative expression in jejunum of antibiotic group and experimental group II was significantly increased(P< 0.05).High-throughput sequencing of bacterial16 Sr DNA of cecum content of broiler showed that the diversity and richness of intestinal microflora(mainly distributed in phylum sclerecta)in broilers were improved by the addition of Calyx Seu Fructus Physalis ultramicro-pulverised powder.Compared with the control group,95% of the treatment groups have improved the ratio of Lachnospiraceae and Lachnoclostridium in intestinal flora.The result shows that Calyx Seu Fructus Physalis ultramicro-pulverised powder increases the villi height and villi height /crypt depth of jejunum and ileum and decreases crypt depth of jejunum,increases Occludin m RNA relative expression,improves the diversity and richness of intestinal beneficial bacteria,adjust the composition and structure of intestinal microbes,thereby improving the intestinal function of broilers.Conclusions: 1.Calyx Seu Fructus Physalis ultramicro-pulverised powder improves growth performance immunity and oxidation resistance of broilers(the optimal dose is 0.32%);2.It improves intestinal mucosa structure and increase tight junction protein Occludin m RNA relative expression of jejunum;3.Improves the diversity and richness of intestinal beneficial bacteria,adjust the composition and structure of intestinal microbes,thereby improving the intestinal function of broilers.
