Objective: Adipogenesis is closely related to obesity,and lnc-ORA is an important potential target for regulating adipogenesis.In addition,aerobic exercise is one of the best ways to combat obesity.Therefore,experiments were designed to investigate the potential mechanism of aerobic exercise suppressing fat deposition through regulating lnc-ORA.Methods: The eight weeks old SPF male mice were non-artificially classified into three groups: normal control group(NC group),high-fat diet group(H group),and high-fat diet plus exercise group(HE group).Among them,the NC group adopted a standard diet,while the H group adopted a high-fat diet to construct an obesity model.And the HE group received eight weeks of aerobic exercise intervention on the basis of a high-fat diet.Aerobic exercise intensity: 18m/min×45min,6% slope,6 days/week.The weight and length were measured once a week.And after the last exercise,2ml blood sample was taken from the eye socket,glucose and insulin tolerance tests were completed,and adipose tissue was dissected and weighed.Plasma was separated and blood lipid levels were detected by spectrophotometer.HE staining of adipose tissue was used to observe the changes of cell morphology.The gene levels of lnc-ORA,FASN,PPARγ,FABP4 and TNF-α in adipose tissue were detected by q PCR.The protein levels of p-PI3 K,p-AKT,p-m TOR,PPARγ,FABP4,FASN,INSR,GLUT4 and p-IRS1 were measured by Western blot.Results:(1)At the end of the experiment,body weight,adipose tissue weight,and lee’s index of mice in the H group were significantly increased compared with those in the NC group(P<0.05),and 8 weeks of aerobic exercise intervention significantly improved these physiological changes(P<0.05).(2)Compared with the NC group,the content of TC and LDL-C was observed to be markedly up-regulated in the H group,and the content of HDL-C was also obviously up-regulated(P<0.05).But after 8 weeks of aerobic exercise intervention,blood lipid levels in mice in the HE group were distinctly improved(P<0.05).(3)The expression level of the adipogenic marker gene PPARγ,FASN and FABP4(P<0.05),and the size of adipose cells were obviously up-regulated in the H group compared with the NC group.However,after 8weeks of aerobic exercise intervention,the expression of PPARγ,FASN and FABP4 was significantly down-regulated(P<0.05),and the size of adipose cells were significantly reduced in HE group.(4)The H group mice developed insulin resistance,which down-regulated the phosphorylation levels of the PI3K/AKT/m TOR pathway(P<0.05).After 8 weeks of aerobic exercise intervention,the expression level of lnc-ORA was down-regulated and the phosphorylation levels of PI3K/ AKT /m TOR was up-regulated in HE group(P<0.05).(5)Compared with the H group,insulin sensitivity of mice was increased and the expression levels of INSR,p-IRS1 and GLUT4 were also significantly up-regulated,while the expression level of TNF-α was obviously decreased in the HE group(P<0.05).Conclusion:(1)Aerobic exercise can reduce body weight and lee’s index,improve the level of blood lipid,and inhibit fat deposition.(2)13 weeks of high-fat diet feeding caused insulin resistance in mice,which eliminated the activation effect of lnc-ORA on the PI3K/AKT pathway.(3)Aerobic exercise can improve insulin resistance through the IRS1/PI3K/AKT/GLUT4 pathway.(4)Aerobic exercise can inhibit PPARγ-mediated adipogenic differentiation by regulating lnc-ORA,thereby inhibiting adipose deposition. |
