Experime ntal purposes: diabetes is one of the most common chronic metabolic disease,the lifestyles of population aging,urbanization developing,city environmental pollution,food crisis,unhealthy diet,obesity and sedentary lead to diabetes mellitus.Type II diabetes is mainly caused by IR resistance,while there is no effective treatment at present,so to improve the cognition of type II diabetes and pay attention to early treatment,meanwhile seek an effective method for the prevention and treatment of type II diabetes is very significant.The experiment study the effects of PI3K/Akt/GLUT4 pathway,and what better intensity exercise can improve the rat myocardial cells of PI3 K,and Akt protein content and promote GLUT4 gene expression,to reveal the movement mechanism of Ⅱ diabetes treatment,exercise therapy for Ⅱ diabetes patients provide theoretical basis for exercise intensity.Experime ntal methods: Experiments with 45 adult male Wistar rats as the research object,and randomly assigned to normal control group o f 8,the remaining 37 divided into diabetic group,preparing Ⅱ diabetes rats model building.Diabetic rats were fed with high fat and high sugar diet(10% sucrose,10% lard,and 5% cholesterol),after 4W then fasting 12 h,intraperitoneal injection of streptozotocin(STZ)to establish a diabetic rat model.To modeling were randomly divided into 32 rats to the success of diabetic control group(DM group,n=8),diabetic low-intensity sport group(DLS group,n=8),diabetic medium intensity sport group(DMS group,n=8),diabetic high intensity sport group(DHS group,n=8).The training scheme is based on the Bedford program while improve the different intensity of rat treadmill movement,C group rats do not exercise,free living feeding in the cage.The exercise group rats on treadmill after adaptive training,the rats in group DLS at 15 m/min speed of 30 min per day;the rats in group DMS at 25 m/min speed of 30 min per day;the rats in group DHS began moving at a speed of 25 m/min,20 min on the first day,and then with 1 m/min increased the treadmill speed,until 30 m/min termination.Each rat rest 1D within 6D,totally exercise 6w.After training and fasting,decapitated.Blood glucose test using blood glucose meter.To measure the serum insulin content by insulin radioimmunoassay kit,measured AKT protein content of PI3 K,ELISA kit,by real-time fluorescence quantitative PCR method for the determination of GLUT4 in rat myocardium gene expression in muscle.Adopting the glycogen kit to determination the rats of myocardial glycogen content.Experime ntal results:(1)After modeling,the diabetes group compared with the normal control group,diabetes group weight gain slow,or even lower.Increased blood glucose no less than 16.7mmol/L.Combined with the behavioral characteristics of multiple feces,polyuria,polydipsia and overeating,the success of type II diabetes mellitus can be determined.(2)Compared with diabetic control group,diabetic exercise group blood glucose,serum insulin levels were decreased.Among them,the middle and high intensity group serum insulin level decreased significantly(p≤0.05),low-intensity exercise group decreased significantly(p≤0.01).The blood glucose levels of rats in each exercise group were significant difference(p≤0.01).(3)Compared with diabetic control group,the body weight,insulin sensitivity index,PI3 K protein content,AKT protein content,GLUT4 gene expression and myocardial glycogen content of diabetic exercise group were all increased.Among them,the body weight,insulin sensitivity,GLUT4 gene expression in moderate and high intensity exercise group were significant difference(p≤0.05),low intensity exercise group was extremely significant difference(p≤0.01).Low intensity exercise group PI3 K protein content,glycogen content has extr elemy significant difference(p≤0.01),high intensity exercise group has significant difference(p≤0.05).Three groups of AKT protein content have very significant difference(p≤0.01).(4)Compared with diabetic group,low intensity exercise group had very significant difference(p≤0.01).Experime ntal conclusions:(1)By different intensity treadmill exercise in diabetic rats blood glucose,serum insulin decreased to different extent,improves insulin sensitivity,improve PI3K/AKT signal pathway of PI3 K and AKT protein content,increased GLUT4 gene expression and on glucose transport capacity.(2)The different intensity runners have increased the sensitivity of the heart muscle with the PI3K/AKT/GLUT4 signaling pathway,alleviate the insulin resistance,and has a good curative effect on type II diabetes mellitus.(3)In the three kinds of exercise inte nsity,low intensity exercise has the most obvious effect on the indexes of myocardium of type II diabetic diabetic rats,and has better effect on improving insulin resistance. |
