Background:The previous studies have confirmed the long-term planting of Fusobacterium nucleatum(Fn),which can reshape tumor microenvironment,and weaken the anti-tumor immune response of the body,thereby promoting malignant proliferation of esophageal squamous cell carcinoma(ESCC)cells,significantly shortening the patient’s survival.In previous studies,the relationship between Fn infection and the age of ESCC patients was not clear.Therefore,in this study,ESCC patients of lower age group(≤50 years old)and higher age group(≥75 years old)were selected as the research objects,and Fn colonization in paraffin-embedded carcinoma tissue samples after surgical resection was detected,and the effects of Fn infection on the survival of ESCC patients of lower age group and higher age group were analyzed.In order to provide reference for early warning and prognosis improvement of ESCC.Objective:To analyze the difference of Fn infection rate in cancer tissue of ESCC patients between the younger age group and the older age group and the effect of Fn infection on the survival time of the two groups of patients,and to explore its clinical significance.Methods:1.The clinicopathological information of 141 ESCC patients undergoing surgical resection in Anyang Cancer Hospital was retrospectively analyzed.64 ESCC patients in the lower age group(≤50 years old)and 77 ESCC patients in the higher age group(≥75years old)were enrolled.2.Fn colonization in cancer tissues was detected by RNAscope and Q-PCR.3.Chi-square test was used to analyze the correlation between Fn infection and clinicopathological features of ESCC patients.4.COX regression was used to analyze the influence of various factors on the prognosis of ESCC patients in low and high age groups.5.Kaplan-meier method was used to draw the survival curve,and log-rank was used to analyze the difference of survival time.6.SPSS 26.0 software was used for data analysis,and the test level α=0.05,P<0.05 was considered as difference.Results:1.The RNAscope and Q-PCR results of Fn infection showed that the positive rate of Fn infection was 44.68% and 46.81%,respectively,and the two detection results were significantly consistent(Kappa>0.7).2.The positive rate of Fn infection in cancer tissues of ESCC patients in the high age group aged ≥75 years was significantly higher than that of ESCC patients in the low age group aged ≤50 years(P<0.05).3.ESCC patients with positive Fn infection are mostly male patients with smoking,drinking and positive family history,and most of them have early lymph node metastasis and clinicopathological stage III/IV(P<0.05).4.The depth of tumor invasion was an independent risk factor affecting the prognosis of ESCC patients in low and high age groups(P<0.05),while Fn infection was only an independent risk factor affecting the prognosis of elderly ESCC patients(P<0.05).5.Fn positive ESCC patients in low age group and high age group had shorter survival,but only the high age group had significantly shorter survival(P<0.05).Conclusions:1.The Fn infection rate of ESCC patients in the older age group was significantly higher than that in the younger age group,and the prognosis was worse.2.Male,smoking,drinking,and family history-positive ESCC patients are positively associated with Fn infection.
