Background:Fusobacterium nucleatum(Fn)is a gram-negative spore free anaerobic bacterium. It is a conditional pathogen and is often found in the mouth, genitals, gastrointestinal tract and upper respiratory tract. Many studies have shown that Fn is closely related to the occurrence and development of colorectal cancer(CRC),gastric cancer, oral cancer and other tumors. Some reports based on detection techniques such as immunohistochemistry and gene amplification show that Fn infection may also be related to esophageal cancer.A research team in Japan reported that the content of deoxyribonucleic acid(DNA)of Fn in esophageal cancer tissue is significantly higher than that in matched normal esophageal mucosa. The DNA positive of Fn is related to tumor clinicopathological stage and tumor specific survival rate. The serum levels of Fusobacterium nucleatum Ig A(Fn-Ig A)antibody and Fusobacterium nucleatum IgG(Fn-IgG)antibody in CRC patients were significantly higher than those in healthy controls. The levels of Fn-IgG antibody in serum of poorly differentiated CRC patients were higher than those of highly differentiated CRC patients; It has been reported that the DNA content of Fn in CRC and its precancerous lesions is high, and the corresponding serum Fn-Ig A antibody is also increased, indicating that there is a certain correlation between Fn antibody and Fn infection; In order to further explore the association between Fn infection and esophageal squamous cell carcinoma(ESCC),Fn-IgG antibody in the serum of ESCC patients was detected to confirm the correlation between Fn infection and ESCC.Objective:To study the correlation between Fn-IgG antibody and ESCC,and analyze the correlation between Fn-IgG antibody and clinicopathological features of ESCC patients. Methods:From April 2021 to July 2021,182 serum samples of hospitalized patients with ESCC diagnosed for the first time and without any treatment in the Department of thoracic surgery of Anyang Cancer Hospital Affiliated to Henan University of science and technology were collected as esophageal squamous cell carcinoma group, and 91 serum samples of healthy people who underwent physical examination in the hospital at the same time were collected as control group. The expression of Fn-IgG antibody in esophageal squamous cell carcinoma group and control group was detected by enzyme-linked immunosorbent assay(ELISA).The general clinicopathological characteristics of all ESCC patients were collected and analyzed by spss23.0 software, chi square test was used to compare the positive rate of Fn-IgG antibody between the two groups, and the correlation between Fn-IgG antibody and clinicopathological features of ESCC patients was statistically analyzed.Results:1.Correlation between Fn-IgG antibody expression and clinical characteristics of182 patients with ESCC:the positive rates of Fn-IgG antibody in esophageal squamous cell carcinoma group and control group were 20.9%and 8.8%respectively.The positive rate of Fn-IgG antibody in esophageal squamous cell carcinoma group was higher than that in control group. The difference between the two groups was statistically significant(c~2=6.327,P<0.05).2.Correlation analysis between Fn-IgG antibody and clinical characteristics of182 ESCC patients:the positive rates of Fn-IgG antibody in ESCC patients with smoking history and ESCC patients without smoking history were 31.9%and 14.2%respectively.The positive rate of Fn-IgG antibody in ESCC patients with smoking history was higher than that in ESCC patients without smoking history. The difference between the two groups was statistically significant(c~2=8.147,P<0.05);The positive rates of Fn-IgG antibody in ESCC patients with and without drinking history were35.1%and 14.4%respectively.The positive rate of Fn-IgG antibody in ESCC patients with drinking history was higher than that in ESCC patients without drinking history. The difference between the two groups was statistically significant(c~2=10.142,P<0.05);The positive rates of Fn-IgG antibody in patients with c TNM stage I,II,III and IV ESCC were 12%,12%,21.3%and 40.6%respectively.The positive rate of Fn-IgG antibody in patients with stage I+II,III and IV ESCC increased in turn. The difference between the groups was statistically significant(c~2=11.141,P<0.05).3.Correlation between Fn-IgG antibody expression and postoperative pathological features of 88 patients with ESCC:the Fn-IgG antibody positive rates of patients with poorly differentiated ESCC and patients with highly and moderately differentiated ESCC were 43.8%and 12.5%respectively.The Fn-IgG antibody positive rate of patients with poorly differentiated ESCC was higher than that of patients with highly and moderately differentiated ESCC.The difference between the two groups was statistically significant(c~2=8.594,P<0.05).Conclusion:1.The expression of Fn-IgG antibody was correlated with ESCC.2.The expression of Fn-IgG antibody was correlated with smoking history, drinking history,c TNM stage and differentiation degree of ESCC patients.
