| Object:To explore the effect of different doses of vitamin C(ascorbic acid,AA)on the sur vival of fat transplantation in autologous fat transplantation.Provide a new method to pr omote the survival of fat transplantation for the clinic.Methods:The experimental animals were 20 female SD rats,randomly divided into low-dos e group(group A),high-dose group(group B),super-high-dose(group C)and control gr oup.The abdominal mesenteric fat of each group of rats was taken out,and 0.8 g of the fat particles were transplanted into a 1 cm diameter subcutaneous sac on the back of eac h rat after proper treatment.The experimental groups were intraperitoneally injected wit h 1 ml of vitamin C solution of 50 mg/d,150 mg/d and 1000 mg/d respectively for 7 co nsecutive days after operation.The control group was intraperitoneally injected with the same amount of normal saline.After 8 weeks,the rats were euthanized.Then the grafts were taken out and evaluated as follows after the adherent fibrous connective tissue was removed:(1)general observation;(2)wet weight;(3)HE staining;(4)immunohistoche mical staining of CD34.Results:(1)The texture of fat in the experimental group was generally softer than that of th e control group,and there were more small blood vessels on the surface of the fat.(2)The graft retention weight of experimental group was significant higher than tha t of the control group,and the retention weight of group B had the maximum weight.(C ontrol group 0.31±0.02 g,Group A 0.38±0.02 g,Group B 0.57±0.02 g,group C 0.45±0.02 g,P < 0.001).(3)The formation of vesicles in the experimental group was reduced.Groups B and C were more effective in reducing inflammatory infiltration and fibrosis.(4)The capillary density in group B and C was higher,group B was(9.93±1.49)pe r high-power field,and group C was(9.20±1.15)per high-power field.The capillary de nsity of the two groups was higher than that of the control group(P<0.001).Conclusion:High-dose vitamin C can increase the capillary density and retention quality of aut ologous fat in rats,reduce the degree of inflammatory infiltration and fibrosis,and reduc e the formation of vesicles. |
