| Objective:To analyze the general clinical characteristics of IBS patients and the gene polymorphism of DA gene rs4680,5-HT transporter gene rs4795541,rs2020938 and5-HT2 A receptor gene rs6313 and rs6311.Methods:From March 2020 to March 2022,patients with IBS with abdominal pain,changes in defecation habits and / or defecation frequency and no organic changes in enteroscopy in the outpatient and inpatient department of Gastroenterology of ChinaJapan Union Hospital of Jilin University were selected to fill in the SAS,SDS and IBSSSS,and 5ml of their venous blood was taken,the target DNA was extracted and the primer sequence was designed.The polymorphisms of DA gene rs4680,5-HT transporter gene rs4795541 rs2020938 and 5-HT2 A receptor gene rs6313 rs6311 were analyzed.Group the IBS patients and healthy control group,record their name,age,gender and other basic information,and explore the gene polymorphism of DA gene rs4680,5-HT transporter gene rs4795541 rs2020938 and 5-HT2 A receptor gene rs6313rs6311 in IBS patients and healthy control group.The relationship between IBS and polymorphisms of dopamine gene rs4680,5-HT transporter gene rs4795541 rs2020938 and 5-HT2 A receptor gene rs6313 rs6311 was obtained.Results:1.In IBS group,the proportion of women,anxiety and depression was significantly higher than that in healthy control group,and the age was significantly lower than that in healthy control group;2.In elderly IBS patients,female patients are less than male patients,and in non elderly IBS patients,female patients are more than male patients;3.IBS-C is more common in elderly patients and IBS-D is more common in non elderly patients;4.Among IBS patients,female,non elderly,IBS-D patients and COMT rs4680 GG,SERT rs4795541 SS,5-HT2 A receptor gene rs6313 AA IBS patients had higher IBS-SSS scores;5.The frequency of COMT gene rs4680 GG genotype and G allele,SERT gene rs4795541 SS genotype and "S" allele,5-HT2 A receptor gene rs6313 AA genotype and A allele in IBS group were significantly higher than those in healthy control group;6.The frequency of AA genotype of COMT gene rs4680,SERT gene rs4795541 LL genotype,SERT gene rs2020938 TT genotype and 5-HT2 A receptor gene rs6311 CC genotype in IBS-C group was higher than that in healthy control group;The frequency of COMT gene rs4680 GG genotype,SERT gene rs4795541 SS genotype and 5-HT2 A receptor gene rs6313 AA genotype in IBS-D group were significantly higher than those in healthy control group;The frequency of SERT gene rs2020938 TC genotype and 5-HT2 A receptor gene rs6313 GA genotype in IBS-M group were significantly higher than those in healthy control group;The frequency of 5-HT2 A receptor gene rs6313 AA genotype in IBS-U group was significantly higher than that in healthy control group;7.Non elderly,female,anxiety and depression COMT rs4680 GG and SERT rs4795541 SS were independent risk factors for IBS.Conclusions:1.The incidence of IBS in young people is higher than that in the elderly,and it is more common in women.There was no significant difference between sexes in elderly patients with IBS.2.IBS-C was more common in elderly patients and IBS-D was more common in non elderly patients.3.The incidence of IBS-C was high in COMT rs4680 AA,SERT rs4795541 LL and rs2020938 TT,5-HTR2 A rs6311 CC gene expressors;The incidence of IBS-D was high in COMT rs4680 GG type,SERT rs4795541 SS type,and 5-HTR2 A rs6313 AA gene expressor;The incidence of IBS-M was high in SERT gene rs2020938 TC and 5-HTR2 A rs6313 GA gene expressor;The incidence of IBS-U was high in 5-HTR2 A rs6313 AA gene expressor.4、COMT rs4680 GG type,SERT rs4795541 SS type and 5-HTR2 A rs6313 AA type non elderly female IBS-D patients have relatively severe clinical symptoms.5.Non elderly,female,anxiety,depression,COMT rs4680 GG type and SERT rs4795541 SS type are independent influencing factors of IBS. |
