| Objective Persistent infection with human papillomavirus(HPV)is an essential factor for the tumorigenesis of cervical cancer.HPV infection could be screened by detecting the expression of major capsid protein L1 of HPV.In our study,L1 monoclonal antibodies were used to detect L1 protein of different types of HPV and clinical pathological tissues to verify the detection ability of the monoclonal antibody.This study can provide information for the research and development of HPV screening kits.Methods(1)The HPV monoclonal antibodies were divided into seven groups which were 3A9,5A4,4G12,3A9+5A4,3A9+4G12,5A4+4G12 and 3A9+5A4+4G12 group.The same amount of L1 protein were detected by Western blot with these antibody groups.Types tested included HPV 16,18,31,45,52,and 58.Then the results were compared among the different groups.(2)Different types of L1 were evaluated by SDS-PAGE with Coomassie blue staining and Western blotting.(3)The HPV monoclonal antibodies were divided into seven groups.ELISA was performed to detect the same amount of L1 protein of HPV 18 and 52 with these antibody groups.(4)Immunofluorescence staining was used to detect HPV L1 protein in condyloma acuminate cells and Cervical exfoliated cells.At the same time,the HPV typing kit was used to detect exfoliated cells in these cases.Results(1)Monoclonal antibodies 3A9,5A4 and 4G12 have different ability to detect HPV L1 protein.In the experiment,the antibody group could detect six types of HPV L1 protein.The antibody group has obvious advantage in detecting L1 protein of HPV18 and HPV52.Although it is not the most sensitive group in detecting other types of L1 protein,it shows stable detection ability.(2)The combination of monoclonal antibodies 3A9,5A4 and4G12 has different ability of L1 protein detection of different types.(3)Monoclonal antibodies 3A9,5A4,and 4G12 can be used to detect HPV L1 protein by ELISA,and the combination of antibodies has obvious advantage.(4)The combination of monoclonal antibodies 3A9,5A4,and 4G12 can detect HPV L1 protein in HPV typing positive samples.Conclusion The results preliminary indicated that the combination of monoclonal antibodies 3A9,5A4 and 4G12 could detect L1 protein of different HPV types.Moreover,the combination of three antibodies had better detection ability than one or the combination of two antibodies.For L1 protein of different HPV types,the detection ability of this antibody group was also different.The antibody group can detect HPV L1 protein by ELISA.It can also detect the expression of HPV L1 protein in condyloma acuminatum and cervical exfoliated cells.It could be seen that the monoclonal antibody group has potential in the development of HPV screening kits for prophylactic of cervical cancer. |
