| Objective:To investigate the effects of sinapine thiocyanate on the proliferation,colony formation,migration and invasion of cutaneous squamous cell carcinoma in A431 and Colo-16 cells,as well as its molecular mechanismMethods:CCK-8,colony formation assay,wound healing and transwell assays were performed to detect the effects of different concentrations(0,5,10 and 20μmol·L-1)of sinapine thiocyanate on the proliferation,colony formation,migration and invasion of A431 and Colo-16 cells;Molecular docking technology were used to verify the binding between sinapine thiocyanate and AKT protein;Western blot was used to detect the effects of sinapine thiocyanate on the expression of AKT,p-AKT(S473),β-catenin,N-cadherin,E-cadherin,Vimentin and PCNA in A431and Colo-16 cells.Results:Sinapine thiocyanate inhibited the proliferation,colony formation,migration and invasion of A431 and Colo-16 cells(P all<0.05);The activated pocket of AKT can stably bound with sinapine thiocyanate;Western blot showed that sinapine thiocyanate decreased the expression of p-AKT(S473),β-catenin,N-cadherin,Vimentin and PCNA in A431 and Colo-16 cells(P<0.05),as well as increasing the expression levels of E-cadherin(P<0.05).Conclusions:Sinapine thiocyanate inhibits the proliferation,colony formation,migration and invasion of human cutaneous squamous cell carcinoma A431 and SCL-1 cells via AKT/β-catenin pathway. |
