Objective: This paper mainly observed the effects of Fuyuan Xingnao Decoction on the expression levels of PI3 K,AKT protein,inflammatory factors TNF-α and IL-6 in diabetic rats with cerebral infarction,in order to explore part of the mechanism of Fuyuan Xingnao Decoction in preventing and treating diabetes mellitus with cerebral infarctionMethods: A total of 150 healthy male Wistar and GK rats were selected for SPF.Rats were randomly divided into normal control group(15),the sham(15),the model group(30),the Western medicine group(30),the Chinese medicine group(30),FGF21 group(30).The normal group used Wistar rats to feed,and the remaining groups used GK rats with high sugar feeding,and the MCAO model was prepared after 2 months.Normal group,fake surgical group and model group is equivalent to a physiological saline gas gantry,Western medicine group is equivalent Nimoda + dimethacuanide medicine,the Chinese medicine group is equivalent to a wake-up soup,FGF21 group injection isometric FGF21 The restructuring factor,the specific dose is based on the rat size.The weight change in rats were detected at each point in time,and the nerve defect score was used to detect blood glucose and blood glucose test strips.HE staining observed brain tissue pathological form changes,ELISA method was used to detect serum TC,TG,FGF21 content and inflammatory factor IL-6,TNF-α level,Western blot technology detects PI3 K,Akt protein levels.Results: 1.Changes in body weight of rats:The weight equilibrium distribution of rats in each group before surgery,no significant difference(P >0.05);24 h after heavy treatment of normal group,7 days after surgery,14 days after surgery continued to rise;compared to the fake surgical group,Western medicine group,The mortgage of the Chinese medicine group and the FGF21 group showed a decline(P <0.01);Compared with the model group,7 days and 14 days after surgery,the body weight of the Chinese medicine group was significantly reduced(P <0.01),the western medicine group and the FGF21 group decreased,no statistics Learn the difference.2.Changes in Bederson score: There was no neurological deficit in the sham operation group;the neurological deficit in the model group,western medicine group,traditional Chinese medicine group and FGF21 group was obvious,indicating that the rat MCAO model was successfully established.The neurological score of the traditional Chinese medicine group gradually decreased from 24 h,3d,7d to 14 d after operation,and there was a significant difference compared with the model group(P < 0.05),and the difference in the scores decreased significantly at 7d and 14 d after operation(P<0.05).3.HE staining results: No obvious edema was found in the nerve tissue in the normal group and the sham operation group,and the nerve cell structure was intact.In the model group,the number of neurons decreased,the brain tissue was edematous and inflamed,the gap increased,and a large number of inflammatory cells were infiltrated.Compared with the model group,the brain tissue structure of each treatment group was slightly abnormal,the number of neurons did not decrease,a small number of pericellular spaces increased,and there was no obvious inflammatory cell infiltration in the tissue,and the degree of damage was significantly reduced compared with the model group.4.Results of blood glucose changes in rats after treatment:After administration,the glycemia of the Western medicine group,the Chinese medicine group and the FGF21 group decreased(P <0.05),and the difference in blood glucose decrease in three groups was not obvious.5.ELISA test results: Compared with the normal group,the level of TC in the sham operation group was increased(P <0.05);compared with the sham operation group,the levels of TC and TG in the model group were significantly increased(P <0.01),and the level of TC in the western medicine group was increased(P<0.05);TG in western medicine group and traditional Chinese medicine group was significantly lower than that in model group(P <0.01),TG in FGF21 group was significantly lower than that in model group(P <0.05);TC in traditional Chinese medicine group was significantly lower than that in model group(P <0.01).Compared with the normal group and the sham operation group,the level of TNF-α in the model group was significantly increased(P <0.01),and the western medicine group and FGF21 group were significantly increased(P <0.05),FGF21 group TNF-α level decreased significantly(P <0.01).The levels of IL-6 in the model group and western medicine group were significantly increased compared with those in the sham operation group(P <0.01).The level of IL-6 in the traditional Chinese medicine group was significantly decreased(P <0.05),and the level of IL-6 in the FGF21 group was significantly decreased(P <0.01).The content of FGF21 in the model group was significantly higher than that in the normal group and the sham operation group(P < 0.01),The content of FGF21 in the FGF21 group was significantly higher than that in the sham operation group(P <0.01);the content of FGF21 in the western medicine group and the traditional Chinese medicine group was significantly lower than that in the model group(P <0.01).6.WB method test: Compared with normal group and sham operation group,the model group PI3 K,p-PI3 K protein level decreased(P <0.05),the treatment of PI3 K,p-PI3 K protein levels were significantly increased by the model group(P <0.05).The p-PI3K protein level of Western medicine group was significantly higher than the Chinese medicine group,and the FGF21 group was significantly increased(P <0.05).Compared with the normal group and the sham operation group,the p-Akt protein content decreased(P <0.05);Compared with the model group,the protein content of Akt and p-Akt in the traditional Chinese medicine group,western medicine group were increased after drug intervention(P <0.05),and the protein content of Akt in the FGF21 group was significantly increased(P <0.05);The protein levels of Akt and p-Akt in the traditional Chinese medicine group were significantly higher than those in the western medicine group and the FGF21 group(P <0.05).Conclusion: 1.Fuyuan Xingnao Decoction can improve the neurological function score of diabetic cerebral infarction rats,improve the pathological morphology of brain tissue,improve blood sugar and blood lipid levels;2.The therapeutic effect and neuroprotective mechanism of Fuyuan Xingnao Decoction on diabetic cerebral infarction rats may be related to inhibiting the expression levels of inflammatory factors TNF-α、IL-6 expression level,and up-regulating the expression levels of PI3K and Akt proteins.
