Effects Of Albumin Tannate And Barm Powder On Diarrhea And The Underlying Mechanisms In Juvenile Rats

Background and Objective Albumin tannate and barm powder is a proprietary Chinese medicine based on empirical formulas.It is clinically used to treat diarrhea caused by pediatric enteritis or indigestion,but its mechanism is unknown.In order to study its specific effects and mechanisms,this research studied the therapeutic effect of Albumin tannate and barm powder on diarrhea in juvenile rats and its relationship with the repair of intestinal mucosa.Methods Forty-eight 21-day-old juvenile rats were randomly divided into 6 groups as follows: normal control group,model control group,montmorillonite powder group,albumin tannate and barm powder low-dose,medium-dose and high-dose groups.Except for the normal group,all the other rats were given 4 g/kg/d senna leaf decoction by gavage for 2 weeks.After successful modeling,distilled water,0.8 g/kg montmorillonite powder,0.1 g/kg,0.3 g/kg and 0.9 g/kg albumin tannate and barm powder were respectively given for 5 days,with intragastric administration twice a day.After 5 days,the diarrhea rate and diarrhea index of each group were recorded.And then the animals were killed,the jejunum of rats in each group was stained with hematoxylin-eosin and immunohistochemistry to observe the changes of intestinal mucosa structure and the expression of proliferating nuclear antigen in intestinal mucosa.At the same time,the intestinal mucosa of rats in each group was scraped to determine the content and expression level of protein,DNA,IL-6 and IL-10 respectively.Results After treatment with high dose of albumin tannate and barm powder,the diarrhea of rats was significantly improved,the villus structure of jejunum basically returned to normal,the DNA content of jejunum mucosa was increased,the expression of PCNA was increased,the expression of IL-6 was decreased,and the expression of IL-10 was increased.Medium-dose albumin tannate and barm powder can improve the diarrhea of rats,the expression of IL-6 was decreased,but has no effect on the above indexes.The low dose of albumin tannate and barm powder had no effect.Conclusions Albumin tannate and barm powder has therapeutic effect on diarrhea of juvenile rats and can promote the repair of intestinal mucosa.
