Objective: to establish the model of Severe traumatic brain injury(s TBI)in rats,and to investigate the effect of Xingshen Huoxue Tongfu recipe(XHXTF)on the recovery of brain function of s TBI and its mechanism.Methods: All mice were allotted to three groups randomly: Model Group 10、Control Group 5,、Experimental Group 10.The standard of experimental rats was 9-week-old male SD rats.Each rat weighed between 180 g and 220 g.Establishing STBI rat model by the way of reformation Feeney method.After it was successfully established,to intervene blank control group and model group by NS(normal saline),and experimental group were treated by Huoxue Tongfu recipe(30mg/kg).All SD rats were sacrificed on the 15 th day,using double antibody sandwich Elisa to detect serum interleukin-6(IL-6)and complement C-3(c3)levels,the contents of Erk,p-ERK and Casepase-9 protein were measured by Western blot,the levels of inflammatory cells were detected by HE staining,and the positive rate of Casepase-9 cells was detected by immunohistochemistry.Results: Scores of mNSS of every group: experimental group were higher than the control group(P<0.05),after 7、14 days of intervention respectively Compared with the model group,the scores of the experimental group on the 7th and 14 th day of drug intervention were significantly lower than those of the model group(P < 0.05).The results of open field test in rats were as follows: the total distance of movement(cm),the times of crossing and standing in the experimental group were less than those in the control group on the 7th and 14 th days after administration,but more than those in the model group(P < 0.05).Double antibody sandwich ELISA: compared with the control group,the concentration of IL-6 and C3 in serum of the experimental group were significantly increased(P< 0.05),comparing with the model group,the concentration of IL-6 and C3 in serum of experimental group were sharply decreased(P< 0.05),this is show that the two groups were different truly.Western blot 、immunohistochemistry: comparing with comparion group,ERK,p-ERK,Casepase-9 protein content and Caspase-9 positive cell rate in experimental group were increased.Compared with the model group,the levels of ERK,p-ERK,Casepase-9 and the positive cell rate of Caspase-9 in the experimental group were increased,but decreased.The results of HE staining showed that the number of inflammatory cells in the slices experimental group’s mice brain tissue were significantly higher than control group’s mice,markedly lower than model group’s mice.The water content of brain tissue in the experimental group was significantly lower than that in the model groupConclusion: Ⅰ.The improved Feeney method can successfully construct rat STBI model with good repeatability and controllability.ⅱ.the motor function of s TBI rats were improved Xingshen Huoxue Tongfu recipe effectively,effectively reduce the score of behavior,and increase the exercise distance,the times of traversing and the times of standing.ⅲ.Xingshen huoxue Tongfu decoction can obviously reduce the inflammatory reaction of brain tissue and body induced by s TBI,and reduce the concentration of IL-6 and C3 in serum,the levels of ERK,p-ERK,Casepase-9 protein,Caspase-9 positive cells and inflammatory cells were decreased.ⅳ.Xingshen Huoxue Tongfu Fang can effectively reduce brain edema caused by STBI,reduce brain water content.
