| Angelica is the root of Angelica sinensis(Oliv)Diels,Licorice is the roots and rhizomes of the legume licorice,Glycyrrhiza inflata Bat.or Glycyrrhiza glabra L.,Flavonoids are one of the main effective ingredients of angelica and licorice.They have strong antioxidant activity,scavenging oxygen free radicals,and antibacterial effects,which have strong antioxidant activity,oxygen free radical scavenging and antibacterial effects,and are used in the treatment of anti-inflammatory and analgesic injuries and burns.Scalding in Chinese medicine belongs to the category of “water scalding”,“soup splash injury”,etc.It is one of the more common body injury diseases in clinical practice and one of the most common and extremely complex trauma diseases in daily work and life;The incidence of burns is extremely high,with about 20 million people suffering from various degrees of burns each year.This article conducts scientific research on the effect of flavonoids in angelica and roasted licorice on the treatment of scalds.First of all,compare the vacuum rinsing machine smothering licorice herbs with traditional soaking and drenching methods,and preparation of licorice tablets.Meanwhile,the effects of honey addition,smothering time and drying temperature on the concoction process of honey roasted licorice were investigated based on response surface design method Determination of glycyrrhizin,glycyrrhetinic acid and leachate content to optimize the honey roasted licorice concoction process.The results show that: Softening licorice herbs by using vacuum rinse machine has the advantages of low water content of the tablets and good softening effect.Using the Box-Behnken response surface method,the optimal process is: 25% honey mixing,5h mulling time,70 ℃ drying temperature.Using the optimized process conditions,the glycyrrhizin content was 0.54%,glycyrrhetinic acid content was 2.02%,and the leachate content was 49%,with a yield of more than 80%,which was higher than the traditional concoction process.Secondly,the flavonoids in the herbs were extracted by ethanol heating reflux method,determination of total flavonoid content in medicinal herbs by UV spectrophotometry,Superoxide anion scavenging assay was performed to evaluate the anti-inflammatory activity and compounding ratio of flavonoid components in roasted licorice and angelica,to prepare three common scalding dosage forms,and to establish a deep II degree scalding model in mice,topical treatment of the wound with different types of agents,and the water content of mouse tissues and wound healing rate were measured at different periods of scald,and the contents of TNF-α,IL-1 in serum and EGF and VEGF in tissues were measured,and conduct slice observation of skin tissue.Experiments show that when the ratio of roasted licorice and angelica is 1:2,the scavenging effect on superoxide anions is the strongest;the three forms have different degrees of therapeutic effects on burns,among which the gelling agent is cool and easy to apply when used,and can form a protective film with the wound to prevent bacterial infection,and shorten the time of the inflammatory storm,which is conducive to the healing of the wound and is the best form for the treatment of burnsFinally,establish the quality standard of the gelling agent and check the physicochemical properties of the gelling agent,the active ingredients in the gelling agent were identified by thin layer chromatography and determined by UV spectrophotometry,examination of the antibacterial effect of gelling agent using the plate coating method.The results showed that the gelling agent had a good antibacterial effect.The obtained gel samples were all brownish yellow gel-like paste.Using ultraviolet spectrophotometry to analyze the main components in the sample,the method is reliable and the repeatability is good.In the accelerated test and stability test,properties,p H,etc.meet the standards of the Chinese Pharmacopoeia 2020 edition,and the effective ingredients do not change much over time,indicating that the gelling agent is stable in nature. |
