Chronic obstructive pulmonary disease(COPD)is the fourth leading cause of death in the world.Traditional Chinese medicine recorded that Brassica napiformis has a high medical and health value,and can prevent inflammation in the lungs,clear sputum accumulation in the trachea,improve airflow obstruction.In this paper,Brassica napiformis ultrafine powder(BUP)was obtained from Brassica napiformis roots,and its physicochemical properties were studied.The COPD mouse model was established by intranasal instillation of Lipopolysaccharide(LPS)to study the effects of BUP on COPD mice.The main research contents and results are as follows:1.Preparation and physicochemical properties of BUP(1)Preparation conditions of BUP: Taking the particle size as t he evaluation standard,the processing conditions of BUP were determined as follows: the loading volume was 0.6 L,the grinding time was 20 min,and the particle size of BUP was 18.311 ± 0.008 μm.(2)Physical parameters of BUP: Compared with ordinary powd er(BOP:passing through 60 mesh sieve after crushing),BUP had smaller apparent structure particles,tended to be spherical,weaker water and oil holding capacity,smaller sliding angle and repose angle,but better storage capacity.(3)Functional factors of BUP: The dissolution rate of functional factors of BUP such as soluble dietary fiber,polyphenols,flavonoids,glucosinolates and isothiocyanates was higher than that of BOP.2.Study on the improvement of COPD mice by BUP(1)Changes in the pathological indexes of COPD mice: BUP could improve the weight loss,lung coefficient increase,emphysema and pulmonary fibrosis of COPD mice.(2)Changes in the inflammatory response of COPD mice: BUP could increase the proportion of regulatory T cells in BALF and blood in mice,and reduced the proportion of dendritic cells,macrophages and neutrophils.BUP could also reduce the levels of inflammatory cytokines such as IL-1 and IL-6 in lung tissue,BALF and serum,and increased the level of an ti-inflammatory cytokine IL-10.Western blot analysis showed that BUP could inhibit the activitation of inflammatory signaling pathway via preventing the dissociation of IκB and repressing p65 in lung tissue and inhibited p65 to release inflammatory factor s.(3)Changes in the oxidative stress in COPD mice: BUP could balance the levels of BALF and reactive oxygen species in blood of COPD mice.BUP could also improve the levels of oxidative stress related factors(such as NO,MDA,SOD,GSH-Px and CAT)in lung tissue of mice.Western blot analysis showed that BUP could promote the expression of Nrf2,HO-1 and NQO-1 in lung tissue to improve the oxidative stress signaling pathway of COPD mice.In conclusion,BUP had a high dissolution rate of bioactive componen ts and activity to improve COPD mice.BUP played an anti-inflammatory role by improving the pathological indexes of COPD mice,regulating the proportion of inflammatory cells,the level of inflammatory cytokines and inflammatory signaling pathway in COPD mice,and played an antioxidant role by regulating the balance of reactive oxygen species,the level of oxidative stress-related factors and oxidative stress signaling pathway in COPD mice.
