| Auricularia auricular has a variety of active ingredients and a wide range of physiological functions.The properties and effects of auricularia auricular products can be affected by many factors including their cell breakage rate(crush size),the solubility of ingredient and the dispersion of powders.Ultrafine grinding technology can increase the dissolution and release of the active ingredient with a high broken rate.However,the dispersion and stability of auricularia auricular ultrafine powders are difficult to improve,β-cyclodextrin(P-CD)is widely used in medicine and food field due to its excellent performance of inclusion.This article explored a new method to prepare auricularia auricular products,not only ensuring a high breakage rate,but providing a kind of product with good water solubility,stability,dispersion and pharmacy property.After preparing auricularia auricular coarse powder(called coarse powder)by mechanical method,we smashed coarse powder by ultra-fine grinding technology and obtained auricularia auricular ultrafine powders(called ultrafine powders).The possibility of inclusion between β-CD and the active ingredient of auricularia auricular in water was investigated by UV spectrum and its apparent inclusion constant was determined with multi-wavelength method.The results showed that the active ingredients of auricularia auricular generated significant inclusion with β-CD as soon as they dissolved out in water and the finer the powder particle size is,the greater the apparent inclusion constant will be,indicating that ultrafine grinding is not only beneficial to the dissolution of active ingredients,but also contribute to the inclusion of the active ingredients.The auricularia auricular/β-CD superfine powders(called superfine powders)were prepared by high shear force mixing method.Then ultrafine powders and coarse powders were identified by DTA/TG,the results of which showed that β-CD can improve the stability of ultrafine powder.The measurement results of solubility and stability showed that the adding of β-CD can increase the solubility of water-soluble active ingredients,while the stability in high temperature and humidity have also been significantly improved.Regard dispersion and clarity as indicators,we investigated the effect of β-CD on the nature of the solution,and measured the change of molecular in the solution by ITC.The results showed that the addition of β-CD can significantly improve the dispersion of ultrafine powder.When we further measured the solution viscosity we found that after addingβ-CD,the dissolution of polysaccharide increased,but the dispersion was improved,and the improvement degree is directly related to the decrease of the viscosity degree of the solution,and the decrease of viscosity is directly related to the significant decrease of the action between polysaccharide molecular caused by β-CD.Auricularia auricular granula was prepared with superfine powder by dry method,which can meet the relative quality inspection requests in particle size uniformity,hardness,hygroscopicity,mobility,suspension,taste and the content of heavy metal,what’s more,the anti-mildew ability of granula was significantly stronger than the coarse powder.Further we mixed superfine powders with common adjuvant to obtain auricularia auricular tablets,which not only have good dispersion characteristic,but also can meet the conventional requests of preparations in appearance,variation of tablets weight,tablets hardness,disintegration,etc.This paper successfully explored a new method for preparing auricularia auricular products,ultrafine grinding technology combined with the inclusion technology to improve products property.This study provided a technology for the industrial production which can simplify the operating process,improve products property and effectively reduce the cost,and also provided new ideas and new methods to research and develop the precious mushroom resource of auricularia auricular. |
