| Objective: To investigate the therapeutic effect of rat gingival mesenchymal stem cells(rGMSCs)and quercetin on periodontitis by local injection into the gingiva of experimental periodontitis model in rats,so as to provide a new idea for minimally invasive periodontal regeneration for the time to come.Methods: Fifty male SD rats were randomly divided into five groups:normal group(group A),periodontitis group(group B),medium group(group C),rGMSCs treatment group(group D)and rGMSCs + quercetin treatment group(group E),with 10 rats in each group.Except for the normal group,the periodontitis models of bilateral maxillary first molars were established in all rats.Six weeks later,one rat in each group was randomly killed to identify whether the model was successful.Group C was injected with 15 μL α-MEM solution in the mesial,central and distal of buccal and palatal side local gingival tissue.Group D was injected with rGMSCs suspension.Group E was injected with rGMSCs suspension containing 10 μM quercetin.After 4 weeks,the rats were killed.Serum and maxillary specimens were collected,and alveolar bone resorption value was calculated.Periodontal tissue sections were made and stained with HE.Serum osteocalcin(OCN)and alkaline phosphatase(ALP)detected by enzyme linked immunosorbent assay(ELISA).Results:1.Six weeks later,plaque accumulation,gingival swelling and other symptoms were found in group B,C,D and E.Histological observation showed attachment loss and inflammatory cell infiltration in periodontal connective tissue.Alveolar bone resorption was observed under stereomicroscope.Periodontitis model was established successfully.2.HE staining results showed that there was no obvious loss of attachment and inflammatory cell infiltration in group A.The migration of combined epithelium to the root side was more obvious in group B,with a large number of inflammatory cells infiltration.Compared with group B,group D and E had less migration of combined epithelium to the root side and infiltration of inflammatory cells.There was no significant difference between group D and E,group B and C.3.The level of alveolar bone resorption in group B was dramatically higher than that in group A(P < 0.05),but there was no dramatical difference between group B and group C(P > 0.05).The level of alveolar bone resorption in group D and group E was significantly lower than that in group B(P <0.05),but higher than that in group A(P < 0.05),and there was no dramatical difference between group D and group E(P > 0.05).4.The content of OCN in group B was higher than that in group A(P <0.05),but there was no dramatical difference between group B and group C(P > 0.05).The content of OCN in group D and group E was significantly lower than that in group B(P < 0.05),but higher than that in group A(P <0.05),and there was no dramatical difference between group D and group E(P > 0.05).5.The content of ALP in group B was higher than that in group A(P <0.05),but there was no dramatical difference between group B and group C(P > 0.05).The content of ALP in group D and group E was significantly lower than that in group B(P < 0.05),but higher than that in group A(P < 0.05),and there was no dramatical difference between group D and group E(P > 0.05).Conclusions:1.The experimental periodontitis model of rats can be successfully established by local injection of endotoxin,gingival separation,ligation of ligation wire and feeding with high glucose and viscous diet.2.Local injection of rGMSCs has anti-inflammatory effect.The content of serum OCN and ALP were decreased,and alveolar bone resorption was inhibited.It has a certain therapeutic effect on periodontitis in rats.3.Quercetin had no effect on rGMSCs in the treatment of periodontitis. |
