| Objective: To investigate the effect of stretch stress stimulation caused by functional position external fixation on the expression of FAK and AKT related genes after acute Achilles tendon injury in rabbits.Compared with the traditional ankle extension(plantar flexion)external fixation after Achilles tendon injury,it aims to provide new ideas for the treatment of acute Achilles tendon injury in the future.Methods: The model of "acute Achilles tendon rupture repair" in adult male Japanese white rabbits was constructed.Postoperatively,the ankle joint was externally fixed in functional position with plaster(group A)and ankle joint was externally fixed in position with extension(group B).At the same time,a sham operation group(group C)was established that only cut the skin and did not damage the Achilles tendon.Achilles tendon tissue was taken 1,3,7,14,28 days after repair of Achilles tendon rupture.We tested the maximum load of Achilles tendon at each time point,and adopted QPCR、Western Blot method to detect FAK,AKT gene expression levels in tendon tissue.Use SPSS22.0 to perform statistical analysis on the obtained data.Results: There was no significant difference in the maximum load of Achilles tendon between the two groups at 1 day and 3 days after operation(P > 0.05);At 7d,14 d and28d,the maximum load of Achilles tendon in group A was higher than that in group B(P < 0.05);The maximum load of Achilles tendon in AB group increased with time.RT q PCR results: On the first day after operation,there was no significant difference in the relative m RNA expression between group A,group B and group C(P > 0.05);On the third day after operation,the relative expression of FAK m RNA in group A was higher than that in group B(P < 0.05),but there was no significant difference in the relative expression of AKT2 and Akt3 m RNA between group A and group B(P > 0.05);The relative expression levels of FAK,AKT2 and Akt3 m RNA in group A were significantly higher than those in group B at 7d,14 d and 28d(P <0.05).The relative m RNA expression of each gene in group A and group B was higher than that in group C except 1 day after operation(P < 0.05).WB results showed that the relative protein expression trend of each gene was consistent with the relative m RNA expression trend of each gene in PCR results.Conclusion:(1)The effect of continuous tension stimulation produced by functional external fixation on Achilles tendon repair was positively correlated with the expression of genes related to FAk-AKT pathway.(2)After repair of rabbit Achilles tendon injury,the tendon healing strength of functional external fixation is higher than that of traditional extension external fixation. |
