Objective:To treat rabbit Achilles tendon rupture with autologous platelet rich plasma(PRP),observe the repair of Achilles tendon morphological structure,the expression of growth factors and the recovery of biomechanical properties,and explore the effects of different concentrations of PRP on tendon healing.Provide a theoretical basis for the clinical practice of PRP application in tendon repair.Methods:48 healthy adult male New Zealand white rabbits weighing 2.5-3.0 kg were taken from the central aorta of rabbit ears.PRP was prepared by the second centrifugation method of Landesberg,and the supernatant was used as a diluent.40%,70%,100%PRP;platelet counts for whole blood and PRP.The Achilles tendon rupture model was prepared by cutting the right hind limb of the rabbit with the Achilles tendon.The rats were randomly divided into the experimental group(Group A,Group B,Group C)and the control group(Group D),with 12 rats in each group.Experimental group:40%,70%and 100%PRP were applied to the A,B and C rabbits respectively after the Achilles tendon was cut off.The control group:the Achilles tendon was directly sutured.The general conditions of the experimental animals were observed after operation.The rabbits were euthanized at 2 and 4 weeks after operation.The Achilles tendon specimens were observed,histologically and immunohistochemically observed,and fibroblast count,collagen fiber content and TGF-?1 were performed,and finally biomechanical detection.Results:The experimental animals survived until the experiment was completed and the incision healed well.Histological observation showed that HE staining:2 weeks after surgery,there was significant difference between the B and C groups and the control group(P<0.05),and the C group was higher than the B group(P<0.05).There was no significant difference in the fibrocyte counts between group A and control group(P>0.05).There were significant differences in the fibrocyte counts between group C and control group at 4 weeks postoperatively(P<0.05).There was no significant difference in fiber cell count between group A and control group(P>0.05).Masson staining:2 weeks and 4 weeks after operation,the difference of collagen fiber content between group B and group C was statistically significant(P<0.05),and group C was higher than group B(P<0.05).Group A Compared with the control group,the difference was not statistically significant(P>0.05).Immunohistochemical staining showed that the expression of TGF-?1 in group B and C was significantly different from that in control group at 2 weeks after operation(P<0.05),and group C was higher than group B(P<0.05).There was no significant difference in the expression of TGF-?1 between group A and control group(P>0.05).There was no significant difference in the expression of TGF-?1 between the experimental group and the control group at 4 weeks after operation(P>0.05).Biomechanical test:The maximum tensile strength of group C was higher than that of the control group at 4 weeks after operation(P<0.05).The maximum tensile strength of group A and B was not statistically significant compared with the control group.(P>0.05).Conclusion:PRP can promote the repair of Achilles tendon injury and improve the healing quality of rabbits.The effect of PRP is 100%.
