| Objective:In this experiment,the liquid processing auxiliary material licorice juice was used as the research object.The orthogonal experiment(L934)was used to investigate the influence of different factors on the preparation process of licorice juice.Optimize the best process by multi-index comprehensive scoring method.Based on this,formulate quality control standards for licorice juice used as auxiliary materials.Using licorice juice prepared by the best technology to study on hepatotoxicity of Evodia.To explore the standardization of preparation and quality standards of licorice juice as auxiliary material for processing preliminarily,on this account,to lay the foundation for quality control of licorice juice and licorice juice processed decoction pieces.Methods:1.A UPLC method was established to determine the contents of liquiritin and ammonium glycyrrhizinate in licorice juice,using acetonitrile as mobile phase A and 0.05%phosphoric acid as mobile phase B for gradient elution.Using orthogonal experimental design to investigate the influence of water addition,decoction time,decoction times on the content of liquiritin and ammonium glycyrrhizinate,and obtain the best preparation process of licorice juice by comprehensive evaluation of the extract rate.2.According to the best preparation technology,prepare the licorice juice from 5factories of licorice decoction pieces which met the quality requirements of licorice herbs in the current pharmacopoeia.To identify liquiritin in licorice juice by thin-layer chromatography;to detect ash content,PH value and soluble solids in licorice juice;to determine the content of ammonium glycyrrhizate and liquiritin in licorice juice by ultra-high performance liquid chromatography and methodological investigation.3.Select Evodia,a representative traditional Chinese medicine made from licorice juice,for the experiment.Mice are divided into normal groups,raw Evodia high,medium and low dose groups(6.1g·kg-1,3.05 g·kg-1,0.61 g·kg-1),licorice juice Evodia high,medium and low dose group(7.5 g·kg-1,3.75 g·kg-1,0.75 g·kg-1).After 15 days of continuous intragastric administration,mice are sacrificed to test the activity of ALT and AST in the serum,calculate the liver index,and detect the activity of SOD and the content of MDA in the liver tissue.Results:1.The content of ammonium glycyrrhizinate and liquiritin are attached with a weight of 0.4 and a creaming rate of 0.2.The comprehensive score selects the best preparation process:add 10 times water for licorice pieces,decoct for 1.5 hours,decoct repeatedly for 3 times,and finally combine filtrate.The results of the analysis of variance showed that the biggest influence on the experimental results was the number of decoctions,and the other two factors had little influence.2.The quality standard of licorice juice has been preliminarily determined:Brownish yellow solution that the content of liquiritin≥1.93%and the content of ammonium glycyrrhizinate≥0.20%,PH value is 5.57±0.27,total ash content is less than 5.50%,acid-insoluble ash content is less than 0.54%,soluble solid content is more than 27.50%.And the quality of licorice is different in different regions,and Gansu licorice has the best quality.3.Compared with the blank group(P<0.01 or P<0.05),the hepatosomal index of raw and processed Evodia rutensis was higher,but the hepatosomal index of the product was lower than that of the raw product(P<0.01).Compared with the blank group(P<0.05),the contents of AST and ALT in raw products and products were higher than those in the blank group(P<0.05),indicating that both raw products and products had liver toxicity.However,compared with the raw products(P<0.01 or P<0.05),the levels of AST and ALT in raw products were generally lower.The content of MDA in raw products and products was lower than that in blank group,but the content of MDA in raw products was higher than that in blank group(P<0.01).Compared with blank group(P<0.05),the SOD content of raw product and product was higher than that of blank group.However,compared with raw product(P<0.01 or P<0.05),the SOD level of product was lower overall.Conclusion:Licorice juice is one of the traditional Chinese medicine processing auxiliary materials,and there is no uniform standard for the existing preparation method of licorice juice.This experiment first optimized the preparation process of licorice juice,which is an auxiliary material of traditional Chinese medicine,and the method is simple,feasible and reliable.A more systematic preliminary study on the quality of licorice juice has been done,and quality standards have been formulated,which are expected to be applied to the quality evaluation of licorice juice.Using licorice juice to process raw evodia,and compare its liver toxicity with raw evodia.Among them,the licorice juice produced evodia has the weakest toxicity,and the low-dose group of processed evodia has the weakest toxicity,which shows that the processed product can significantly reduce the liver toxicity of the evodia.To reduce toxin to improve the safety of clinical medication is an effective traditional Chinese medicine processing technology.Therefore,it is proved that licorice juice is an auxiliary material for processing,improve the safety of using medication in clinical,and provide a reference for the research on licorice juice. |
