Effect Of Jiaji Electroacupuncture On NgR Protein Expression In Rats With Total Transected Spinal Cord Injury

Objective:The model of spinal cord injury was established,and the expression of NgR protein in rats with spinal cord injury was investigated by the treatment of jiaji electroacupuncture,observing the histopathological changes of the injured area and the recovery of motor function before and after treatment,to reveal the neuroprotective mechanism of jiaji electroacupuncture in rats with spinal cord injury,and to provide experimental basis and theoretical support for clinical treatment.Method:54 adult female Wistar rats were divided into sham operation group(blank control group),model group and jiaji electroacupuncture group according to random number table method,with 18 rats in each group.To established the model of spinal cord injury in rats with complete transection,in the sham operation group,cutting the skin,separating the fascia and muscle,only the T10 spinal cord was exposed after removing the lamina without transection,directly sutured without any treatment,and routine postope rative care.In both the model group and the jiaji electroacupuncture group,the spinal cord was transected at the T10 level.After the successful preparation of the spinal cord model,the model group received no treatment and was kept alone in a cage and given routine care after surgery.The Jiaji electroacupuncture group was treated with jiaji electroacupuncture after 2h successful modeling for 30 minutes,and then once daily 30 minutes.Rats in the sham operation group and model group were grabbed and binded once a day,30 minutes.BBB score was used to evaluate the motor function of rats in each group on 3d,7d and 14 d after operation,and then extracting spinal cord tissue from the injured area of rats;using Western Blot to detect the expression of NgR protein,and HE staining to observe the pathological changes of spinal cord tissue.Results:1.The BBB score: BBB scores of the sham operation group were all 21 points at 3d,7d and 14d;compared with the sham operation group,the BBB scores of the model group and the jiaji electroacupuncture group decreased at each time point(P < 0.05),and the decrease was most obvious in the model group;compared with model group,jiaji electroacupuncture group after treatment,3 d,7 d,14 d BBB score at all time points higher than model group,difference has statistical significance(P < 0.05);the intra-group comparison showed that the BBB scores of the Jianji electroacupuncture group after 7d and14 d treatment were significantly higher than those after 3d treatment,w ith statistical significance(P < 0.05).2.HE staining results: In the sham operation group,the morphology of neurons and glial cells was normal without necrosis or edema,and can observe the intact spinal cord tissue structure.In the model group,3d after operation,the spinal cord tissue structure was disordered,the number of neuronal necrosis was increased,the glial cells had a significant tendency of proliferation,and there were more inflammatory cells aggregation;on the 7th day after operation,the spinal cord tissue structure was seriously damaged,glial cells,glial fibers and inflammatory cells were increased,and normal neurons were obviously missing;On the 14 th day after the operation,tissue liquefactive necrosis formed a large number of cavities,inflammatory cells infiltrated,glial cells proliferated,and the number of neuronal death decreased.In the Jiaji electroacupuncture group,the number of neurons increased with the prolongation of treatment time,the number of inflammatory cells gra dually decreased,the degree of inflammatory infiltration decreased,and the structure of neurons and spinal cord were repaired.3.Western Blot results: At 3d,7d and 14 d after operation,the expression of NgR protein in the sham operation group was the lowe st,and in the model group and the jiaji electroacupuncture group was higher than the sham operation group.However,the expression of NgR protein in the jiaji electroacupuncture group decreased after treatment.Compared with sham operation group,the expression of NgR protein in model group was significantly increased,the difference was statistically significant(P < 0.05);compared with the model group,the expression of NgR protein in the jiaji electroacupuncture group decreased significantly,the difference was statistically significant(P < 0.05);compared with the 3d jiaji electroacupuncture group,the expression of NgR protein in the 7d and 14 d groups was significantly decreased,the difference was statistically significant(P < 0.05).Conclusions:1.Jiaji electroacupuncture can repair the damaged spinal cord tissue structure of rats,reduce the pathological reaction and play a neuroprotective role.2.Jiaji electroacupuncture can promote axon regeneration and improve neural function and motor function in rats,the mechanism of which may be related to the down-regulation of NgR protein expression in the spinal cord injury area of rats.
