Experimental Study On The Effect Of Jiaji Electroacupuncture Combined With Treadmill Training On Nerve Function In Rats With Spinal Cord Transection Injury

Objective:The expression of NgR in the spinal cord tissue of the injured area was observed by immunofluorescence staining,and the mechanism of effect of Jiaji electroacupuncture combined with treadmill training on the neurological function of rats with spinal cord transection injury was discussed.Methods:72 adult female Wistar rats,aged 11 to 12 weeks and weighing 200 to 220 g,were randomly divided into four groups according to the random number table method: sham-operated group,(group A,18 rats),model group(group B,18 rats),Jiaji electroacupuncture group(group C,18 rats),Jiaji electroacupuncture combined with treadmill training group(group D,18 rats).Group A only exposed T10 segments of spinal cord,without transection,suture wound,no treatment.The model of spinal cord transection injury was established in the other three groups at t 10 segment.Group B received no treatment after successful modeling.Group C and group D received Jiaji electro-acupuncture intervention once 2,4 and 6 hours after successful modeling,and then once a day.Group D received treadmill training once a day from the 3rd day after successful modeling.Six rats in each group were randomly selected on the 3rd,7th and 14 th day,respectively.BBB score was used to evaluate the recovery of hind limb motor function in rats.Then the injured spinal cord tissues of rats were taken out and detected by immunofluorescence staining.The expression of NgR in the injured spinal cord tissues of rats in each group was observed and compared.Results:1.BBB score:(1)The motor function score of rats after SCI was closely related to the intervening time.At 3 days of treatment,the score of BBB was the lowest,the score of 7 days was increased,and the score of 14 days was the highest.(2)The score of rats after SCI is closely related to the intervention methods.The score of BBB in group A did not change significantly before operation and at each time point after operation.Compared with group B,the BBB score of rats in group C is significantly higher,and the difference is statistically significant(P<0.05);Compared with group C,the BBB score of rats in group D is significantly higher,and the difference is statistically significant(P < 0.05).2.Immunofluorescence staining:(1)The expression of NgR was closely related to the intervening time after SCI.The highest expression of NgR was found on the 3rd day,decreased gradually at 7 days,and the lowest on the 14 th day.(2)The expression of NgR in the injured area after SCI was closely related to the intervention methods.The expression of NgR in spinal cord tissue of group A did not change significantly at each time point.Compared with group A,the expression of NgR in spinal cord tissue of group B was significantly increased,and the difference was statistically significant(P < 0.05).Compared with group B,the expression of NgR in spinal cord tissue of group C was significantly decreased,and the difference was statistically significant(P < 0.05).Compared with group C,the expression of NgR in spinal cord tissue of group D was significantly decreased,and the difference was statistically significant(P < 0.05).Conclusion:1.Jiaji electroacupuncture combined with treadmill training can improve the BBB score of rats with spinal cord transection injury and improve the motor function of rats with spinal cord transection injury.2.Jiaji electroacupuncture combined with treadmill training can inhibit the expression of NgR in the injured area of rats with spinal cord transection injury,promote the regeneration of axons and improve the nerve function.
