| Object:By observing the changes in the phase expression of RhoA protein in the transection area of the rat spinal cord and the recovery of the motor function of the hind limbs under the intervention of Jiaji electroacupuncture at different times,and then studying the repairing effect of Jiaji electroacupuncture on the injured spinal cord and Jiaji Part of the mechanism of the time effect of electroacupuncture.Method:108 Wistar adult female rats were divided into sham operation group(A group),model group(B group),2h group(C group),4h group(D group),and 6h group(E group)using random number table method.And 12h group(F group),a total of 6 groups,each with 18 animals.In the experiment,the T10 segment was injured by the total transverse injury modeling method.After the injury,the model group was not treated with intervention,grabbed and bound once a day;2h group(C group),4h group(D group),6h group(Group E),Group 12h(Group F)4 treatment subgroups,2h,4h,6h,12h,4 time points after the successful modeling,began to intervene Jiaji electroacupuncture for treatment,once a day,every time The sham operation group was not injured,only the spinal cord was exposed and sutured,no intervention treatment was given,and the sham operation group was grasped and bound once a day.On 3d,7d,and 14 d after operation,the rats in each group were scored with BBB behavioral scores to quantitatively evaluate the changes in the motor function of the hind limbs of the rats;then Western Blotting was used to detect the expression of axonal regeneration-related inhibitory protein RhoA,and observe Its changes.Results:1.BBB Behavioral Score(1)At 3d,7d and 14 d after operation,compared with group A,the BBB scores of the remaining groups were significantly reduced,the difference was statistically significant(P<0.05);compared with group B,the BBB scores of the four treatment groups of group C,group D,group E,and group F all improved,and the difference was statistically significant(P<0.05).And the BBB scores of rats in each Jiaji electroacupuncture treatment group showed a trend of being the lowest at the 3rd day of treatment,increasing at the 7th day of treatment,and significantly increasing at the 14th day.(2)At 3 days after operation,there was no significant difference among the four groups in group C,D,E and F(P>0.05).At 7 days after operation,compared with group F,The BBB scores of group C,D,and E all increased(P<0.05).At 14 days after operation,compared with group F,the BBB score of group C was significantly higher,and the difference was statistically significant(P<0.05);compared with group F,the BBB scores of group D and group E increased slightly(P>0.05).After 3d,7d,and 14 d,compared with group C,the BBB scores of group D and group E decreased slightly(P>0.05).2.Western Blotting detection(1)At 3d,7d,and 14 d after operation,the expression level of RhoA protein in the spinal cord injury area of rats in each group was higher than that of group A(P<0.05).Compared with group B,group C,group D Group,E group,F group,the expression of RhoA protein in the 4 treatment groups decreased,and the difference was statistically significant(P<0.05).The condition of RhoA protein in each Jiaji electroacupuncture group showed that the expression level of RhoA protein was the highest at the 3rd day of treatment,the expression level of RhoA protein gradually decreased at the 7th day of treatment,and the expression level of RhoA protein decreased significantly at the 14th day of treatment.(2)At 3 and 7 days after operation,the expression levels of RhoA protein in group C,group D,and group E were lower than those in group F,and the difference was statistically significant(P<0.05);On the 14th day,the expression level of RhoA protein in group C was significantly lower than that in group F(P<0.05);compared with group F,the expression of RhoA protein in group D and E was slightly lower(P>0.05).At 3d,7d,and 14d after surgery,there was no significant difference in the expression level of RhoA protein among the C,D and E groups(P>0.05),and all Jiaji electroacupuncture treatment groups showed overall Out of this,the earlier the interventional treatment,the lower the expression level of RhoA protein in the spinal cord injury area.Conclusion:1.Jiaji electroacupuncture can down-regulate the expression level of RhoA protein in the area of spinal cord transection injury in rats,improve the motor function score of the hind limbs of model rats,and promote the regeneration of damaged axons after spinal cord injury.2.Different intervention timings of Jiaji electroacupuncture have different effects on the recovery of damaged spinal cord nerve function,and early intervention is better than late intervention. |
