Effect Of Rictor On Membranous Cell Decidualization And Expression In Human Uterine Decidual Tissue | | Posted on:2020-11-07 | Degree:Master | Type:Thesis | | Country:China | Candidate:L M He | Full Text:PDF | | GTID:2504306533959209 | Subject:Genetics | | Abstract/Summary: | PDF Full Text Request | | Objective:Decidualization of endometrial stromal cells is essential for embryo implantation and successful pregnancy.Abnormalities can lead to infertility and abortion.In addition to the effects of estrogen and progesterone,some cytokines are involved in this process.m TORC2 plays a role in cell proliferation,growth and differentiation.Rictor protein is a specific scaffold protein,and its role in the process of decidualization has not been reported.This study explored the role of Rictor in the decidualization of stromal cells and its impact on spontaneous abortion in order to lay a foundation for the study of spontaneous abortion.Method:1.The mouse endometrial stromal cells were isolated and cultured,and the Rictor gene of mouse endometrial stromal cells was silenced by Si RNA,and the decidualization was induced artificially.2.Seventeen cases of sterile fresh spontaneous abortion decidual tissue specimens were selected as experimental research group(early spontaneous abortion group)for 8-10 weeks of gestation,and 34 cases of normal artificial abortion decidual tissue specimens were selected as normal control group(normal pregnancy group).3.Immunohistochemistry was used to detect the expression of Rictor and SGK1 in decidual tissue.4.Proteins from human uterine decidual tissue and mouse stromal cells were extracted with protein lysate containing protease inhibitor.Tissue protein was quantified by BCA method,and the expression of Rictor and SGK1 protein was detected by Western blotting.5.The total RNA in decidual tissue and mouse stromal cells was extracted by TRIZOL method.The expression of Dtprp m RNA in Rictor,SGK1 m RNA and stromal cells was detected by reverse-transcriptional Real-time PCR.Result:1.After silencing the Rictor gene,the expression levels of Rictor,SGK1 protein and m RNA were significantly decreased,and the difference was statistically significant(P<0.05).2.After silencing the Rictor gene,the expression level of Dtprp m RNA was significantly decreased,and the difference was statistically significant(P<0.05).3.Immunohistochemistry found that Rictor and SGK1 were positively expressed in the decidual tissue of patients with early spontaneous abortion and normal pregnancy.Compared with the normal group,the expression levels of Rictor and SGK1 in the early spontaneous abortion group were significantly lower.4.The total protein expression of Rictor and SGK1 in the early spontaneous abortion group was significantly lower than that in the normal pregnancy group(P < 0.05).5.The expression of Rictor and SGK1 m RNA in the early spontaneous abortion group was significantly lower than that in the normal pregnancy group(P < 0.05).Conclusion:1.Silencing the Rictor gene can inhibit stromal cell decidualization.2.The reduced expression of Rictor and SGK1 may be one of the causes of abortion. | | Keywords/Search Tags: | Human, Rat, Stromal Cell Degeneration, Spontaneous Abortion, Decidual Tissue, Rictor Gene | PDF Full Text Request | Related items |
