The Research On IL-1β Express In Human Decidual Cell Which Effected By The Lipopolysaccharide

Objective:We cultivate human decidual cells in vitro, treat with different concentrations of lipopolysaccharide (LPS), to study the different expression of the inflammatory factor IL-1(3on the decidual cells.Methods:We took the fresh decidua by normal abortion operation,cultured the cells in vitro. Identification of decidual stromal cells by immunofluorescence detection of placental lactogen.When the generation of decidual cells were culture to4-5generation, it was divided into five groups, we added to different concentrations of LPS (Ong/ml,25ng/ml,50ng/ml,100ng/ml,200ng/ml) to the cells. Treated for24hours,(1) Detection of IL-1(3protein expression in decidual cells by confocal Laser-immunofluorescence (2) To detect the IL-1β mRNA expression by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR), the expression analysis of the group without statistical difference.Results:1)Culture the decidual cells successfully. Identification of decidual stromal cells by immunofluorescence detection of placental lactogen,we found the percentage of prolactin positive staining cells were95%.2)Scanning human decidual cells in each group by confocal laser-immunofluorescence, the IL-1β protein expressed in both the cytoplasm and nucleus.The differences expression of IL-1β were significant in the experimental groups compared in the control group after which were treated by LPS,P<0.05, which had statistical significance. With the concentration of LPS gradually increasing,the expression of the IL-1β protein gradually increased.3) Significant difference in the expression of IL-1β mRNA in each experiment compared with the control group after which were treated by different concentrations of LPS,P<0.05, which had statistical significance.With the concentration of LPS gradually increasing,the expression of the IL-1β mRNA gradually increased.Conclusion:1) After acted LPS on human decidual cells, the expression of IL-1β protein was increased. And with the increasing concentrations of LPS, IL-1β protein expression gradually increased.2)After acted LPS on human decidual cells, the expression of IL-1β mRNA was increased. And with the concentration of LPS, the expression of IL-1β mRNA gradually increased.
