Objective:The aim of this study was to invenstigate the differences in the expression of the inflammatory cytokine-induced enzyme: Cycloxygenase-2 in the human decidual cells, which were cultured in vitro and treated by different concentrations of lipopolysaccharide. Then we explored the correlation between the over-expression of COX-2 and the infection of ducidual cells.Method:Collect normal human decidual tissue which from induced abortion in early pregnancy to primary culture cell in vitro. Decidual cells that were cultured 3th -4th generation were treated by different concentrations of LPS (0ng/ml,25ng/ml,50ng/ml,100ng/ml,200ng/ml). After 48 hours:1) Detect the differences in the expression of COX-2 protein in each group decidual cells by Laser scanning confocal microscopy- influorescence.2) Detect the differences in the expression of COX-2 mRNA in each group dscidual cells by semi-quantitative reverse transcription polymerase reaction (RT-PCR). Whether there have statistical different or not.Result:After scanning human decidual cells in each group by Laser scanning confocal microscopy-influorescence, the COX-2 protein expressed in both the cytoplasm and nucleus. The differernces in the expression of COX-2 were significant in the experiment groups compared in the control group after which were treated by LPS, P<0.05, which had statistical significance. With the concentration of LPS gradually increasing, the expression of the COX-2 protein gradually increased.2) Significant differences in the expression of COX-2 mRNA in each experiment compared with the control group after which were treated by different concentrations of LPS, P<0.05, which had statistical significance. With the concentration of LPS gradually increasing, the expression of the COX-2 mRNA gradually increased.Conclusion:1) After LPS acted on human decidual cells, the expression of COX-2 protein was increased. And with the increasing concentrations of LPS, COX-2 protein expression increased gradually.2) After LPS acts on human decidual cells, the expression of COX-2 mRNA was increased. And with the concentration of LPS, the expression of COX-2 mRNA gradually increased.
