| Purpose:The camel is called"the ship of the desert".It has strong resistance to thirst,hunger,and adaptability to harsh weather.It can survive without water and food for a long time.Studies have found that placenta extract has the effects of repairing tissue damage,anti-aging,anti-tumor,and enhancing immunity.In order to avoid waste of placental resources and environmental pollution,this study aims to study the immune enhancement effect and mechanism of camel placental immunomodulatory factors.Methods:This thesis takes Bactrian camel placenta as the research object,and uses a combination of high-speed freezing centrifugation and ultrafiltration to prepare Camel placenta extract(CPE),and detects its basic physical and chemical properties:p H value,protein concentration,Antioxidant capacity and maximum absorption peak,etc.;select healthy Balb/C mice,male and female,and randomly divided into blank control group,cyclophosphamide immunosuppressive model group,thymosin positive control group,CPE high,medium,and low dose groups(1.2 mg/m L,0.6 mg/m L,0.3 mg/m L).While calculating the index of mouse spleen and thymus,the interleukin-2(IL-2)and interleukin-4(IL-2)in mouse plasma were detected by enzyme-linked immunosorbent assay(ELISA),flow cytometry and histopathological methods.-4),γ-interferon(IFN-γ),CD3~+,CD4~+and CD8~+cell levels and the morphological changes of the spleen and thymus.Detect mouse immunoglobulin G(Ig G)and peripheral by ELISA method and flow cytometry The level of CD19~+and CD40~+cells in the blood is used to evaluate the effect of CPE on the immune function of T and B lymphocytes.By analyzing the above experimental results,comprehensively evaluate the immune enhancement effect and mechanism of CPE.Results:(1)The prepared CPE was a neutral,stable,light yellow,clear and transparent solution,which contained nucleic acids and proteins.(2)The protein concentration of CPE measured is 1.2±0.0271 mg/m L,the nucleic acid concentration is 0.252 mg/m L,and the total antioxidant capacity is 1.48±0.0205 m M/1.2 mg/m L.(3)The effect of CPE on the immune function of B lymphocytes:The expression of CD19~+,CD40~+and Ig G levels in the CPE high,medium,and low dose groups were significantly higher than the model control group(P<0.05),and the CPE high,medium,and low dose groups were immunized The organ index was significantly increased(P<0.05),and it was found through histopathological observation that CPE promoted tissue repair in immunosuppressed mice.(4)The effect of CPE on the immune function of T lymphocytes:Compared with the model group,the expression of peripheral blood T lymphocyte subsets CD4~+and CD8~+in the CPE high,medium,and low dose groups were significantly increased(P<0.05),and CPE was high The level of cytokine IL-2 secreted by T lymphocytes in the middle and low dose groups was significantly reduced(P<0.05),and the levels of IL-4 and IFN-γwere significantly increased(P<0.05).Therefore,CPE had a significant effect on mouse T lymphocytes.Immune function has a promoting effect.In the process of increasing the percentage of CD8~+cells and the level of Ig G production,the effect of CPE was significantly higher than that of thymosin(P<0.05).Conclusion:(1)The method of combining high-speed refrigerated centrifugation and ultrafiltration has successfully prepared a relatively stable CPE.(2)Cyclophosphamide has successfully established an immunosuppressive model in mice.CPE may have a repair effect on the immune organ damage of immunosuppressed mice.(3)CPE promotes the humoral immune response of animals by promoting the expression of immune function-related factors of T and B lymphocytes in immunosuppressed mice and the ability of immunosuppressed mice to secrete antibodies.(5)CPE has a strong immune enhancement effect,and its effect is consistent with or more effective than thymosin. |
