Objective: Cancer has become the most difficult disease to cure.In the process of tumor development,the substances secreted by tumor cells and the substances in tumor microenvironment inhibit the body’s immune response,so that tumor cells escape from the body’s immune surveillance,thus causing tumor immune evasion.Many studies have proved that placental immunoregulatory factor can enhance the immune function of the body.Can camel placenta extract(CPE)enhance immunity by influencing the immune escape process of tumor? At present,it has not been decided.Therefore,this study aims to explore the effect of CPE on tumor immune escape process.Methods: Lewis lung cancer cell line was used to establish tumor model in mice by axillary transplantation,and then randomly divided into model group and administration group,8 mice in each group.After 15 days of continuous intraperitoneal injection of CPE(the model group and the blank control group were injected with the same amount of normal saline),the tumor tissues of mice were taken,and the tumor weight was measured,and the tumor inhibition rate was calculated;Pathological sections were made to observe the morphology of tumor tissue and lung tissue;Detection of IFN in mouse serum by ELISA-γ And IL-12 expression level;Fluorescent quantitative PCR(qPCR)and Western blot(WB)were used to detect the mRNA and protein levels of PD-1 and PD-L1 in tumor tissues.Results: Compared with the model group,the tumor inhibition rate of the treatment group was 14.44%;The pathological section showed that the lung tissue morphology of the treatment group was slightly more complete than that of the model group,but there was no significant difference;The results of ELISA showed that the IFN in serum of the treatment group was higher than that of the control group-γ Compared with the blank control group,the contents of IL-12 and IL-12 were increased,and there were significant differences(P <0.05),but there was no significant difference with the model group.QPCR results showed that compared with the model group,the expression level of PD-1 in the treatment group was slightly lower,but the difference was not significant,while the content of PD-L1 in the treatment group was significantly lower than that in the model group(P < 0.05).WB results showed that compared with the model group,the expression levels of PD-1 / PD-L1 in the treatment group were slightly decreased,and the difference was significant(P <0.05).Conclusion: CPE may increase IFN in serum of mice-γ And IL-12;It can reduce the expression of PD-1 / PD-L1 in tumor microenvironment.These results indicate that CPE has a certain inhibitory effect on tumor immune evasion.The results of this study lay a good foundation for the further study of CPE mechanism.
