| Throughing a lot of exploration,it is believed that the chemical components are the material basis of the cold and hot properties of traditional Chinese medicine.For a traditional Chinese medicine with a single component,the property of its chemical component should be the property of the traditional Chinese medicine.The cold and heat properties of single component Chinese medicine are determined by the structure of its chemical components.For mineral drugs,the structure of chemical composition includes elemental composition,valence state,crystalline state,etc.For organic components,the chemical structure includes the framework structure and functional groups of the compound.For single Chinese medicine containing multiple ingredients,each of them has its specific structure and should have its specific cold and heat properties.The composition and proportion of the prescription are fixed,as are the types and mutual proportions of the chemical components contained.Change the composition of prescription or scale,its place contains a component to be able to produce change inevitably,the medicinal property of prescription also can change subsequently.That is to say,the cold and hot properties of the prescription can be adjusted by the properties of the compound medicine flavor and their mutual proportion.Based on this,we speculated that the cold and hot properties of TCM were determined by the cold and hot properties of the chemical components contained in TCM,and the cold and hot properties of the chemical components of TCM were vector additive.So for the cold Chinese medicine,in addition to the cold component,is there any hot component? Does hot Chinese traditional medicine also have cold component? Is there a difference in how hot or cold each component is? The components of cold and heat in traditional Chinese medicine need to be determined by experimental evaluation.In this study,Rheum tanguticum maxim.ex Balf was selected as the research object,and cellular chinese medicine method were used to evaluate the main components of rhubarb,in order to determine the main material basis of the chilling ability of rhubarb.That is to reveal the main components of rhubarb chill phenicity under the guidance of cold and hot properties determination,so as to enrich the modern scientific connotation of TCM property theory,and provide a research basis for further research on the target,mechanism and "attribute-structure relationship" of rhubarb Cold characteristics.To further prove that the chilling and heat properties of TCM are determined by the chilling and heat properties of its chemical components,and the chilling and heat properties of TCM chemical components are vector additive.In this paper,petroleum ether fraction,trichloromethane fraction,ethyl acetate fraction,n-butanol fraction and water fraction were obtained from the water decoction of rhubarb by systematic solvent method.The types and contents of components in different parts of rhubarb were determined by HPLC and mass spectrometry.Results showed the petroleum ether fractions mainly contained emodin(0.16±0.05 μg/g)and emodin methyl ether(1.20±0.02μg/g).The trichloromethane fraction mainly contained cinnamic acid(7.79±0.15 μg/g),chrysophanol(1.63±0.02 μg/g),emodin methyl ether(0.10±0.01 μg/g),emodin(3.69±0.27μg/g),aloe emodin(2.97±0.33 μg/g),and rhein(2.80±0.30 μg/g).The ethyl acetate extract contained gallic acid(86.46±0.56 μg/g),(+)-epicatechin(89.97±1.10 μg/g),(-)-epicatechin(22.53±0.37 μg/g),cinnamic acid(0.80±0.30 μg/g),aloe emodin(0.11±0.10 μg/g),emodin(0.23±0.03 μg/g),emodin methyl ether(0.06±0.03 μg/g);The n-butanol extract contained gallic acid(67.73±1.41 μg/g),(+)-catechin(30.36±0.50 μg/g),(-)-epicatechin(8.14±0.05μg/g),rhufonic acid-8-O-D-β-glucoside(19.04±0.13 μg/g),rhufonic acid(0.40±0.03 μg/g)and emodin methyl ether(0.07±0.02 μg/g).The water part contained gallic acid(24.15±0.18μg/g),(+)-catechin(3.61±1.20 μg/g),rhuodin-8-O-D-β-glucoside(33.16±0.11 μg/g),rhuodin acid(1.13±0.01 μg/g)and emodin methyl ether(0.05±0.01 μg/g).The cold and hot properties of rhubarb water decoction,petroleum ether extract,trichloromethane extract,ethyl acetate extract,n-butanol extract and water extract were evaluated by cellular chinese medicine method.Results showed the decoction of rhubarb was cold,which was consistent with traditional Chinese medicine.The petroleum ether and ethyl acetate fractions of rhubarb were thermal.The trichloromethane part,n-butanol part and water part are cold.It is preliminarily indicated that there are not only cold components but also hot components in rhubarb.The coldness of rhubarb as a whole may be the combined effect of cold and hot components in rhubarb.The cytochemical properties of 12 compounds in rhubarb were evaluated by cellular chinese medicine method.Results showed emodin,chrysophanol,cinnamic acid,catechin,epicatechin and gallic acid were hot.Aloe emodin,rhein acid,emodin methyl ether,rhein8-O-D-β-glucoside,sensenoside A and sensenoside B were cold.It is further indicated that rhubarb contains not only cold components,but also hot components.Through the analysis of the cold and hot components of each of the five parts,it is preliminary that the main components of the cold of rhubarb may be aloe emodin,rhein acid,emodin methyl ether,and rhein acid-8-O-D-β-glucoside.The degree of coldness and heat of each component is different,and its contribution to the overall coldness is different.The trichloromethane fraction showed coldness,but the content of cinnamic acid was the highest.In order to study the trichloromethane part contains the influence of each component to the overall cold sex,on the basis of trichloromethane areas the proportion of each component content,cinnamic acid and aloe emodin by cellular chinese medicine method(3:1),cinnamic acid and rhein(3:1),cinnamic acid and rhubarb phenol(5:1),cinnamic acid and emodin(2:1),cinnamic acid and emodin methyl ether(7-1)attributes of fever.Results showed the coldiness of cinnamic acid and aloe emodin(3:1),the coldiness of cinnamic acid and rhein(3:1),the coldiness of cinnamic acid and chrysophanol(5:1),the coldiness of cinnamic acid and emodin(2:1),and the coldiness of cinnamic acid and emodin methyl ether(7:1).The results showed that aloe emodin and rhein acid were the main components of cold in trichloromethane parts of rhubarb.The degree of coldness and hot of each component is different,and its contribution to the overall coldness is different,showing vector addition.Under the guidance of "Cold diseases with hot drugs,hot diseases with cold drugs",cellular chinese medicine method was used to verify the cold and heat of 10 compounds.Cell-hot syndrome model was established by using Cinnamomum cassia Presl,Piper Nigrum L.,Curculigo orchioides Gaertn.,which were reversed with aloe emodin,rhein acid,emodin methyl ether and rhein acid-8-O-D-β-glucoside,respectively.Coptis chinensis Franch.and Curculigo orchioides Gaertn.were used to construct the cell cold syndrome model,and the thermal(-)-epicatechin,(+)-catechin,gallic acid,cinnamic acid,emodin and chrysophanol were used to reverse the cell cold syndrome.The results showed that all of them were reversed after modeling.Aloe emodin,rhein,emodin methyl ether,rhein 8-O-D-β-glucoside,sennaloside A and sennaloside can realize the "cold medicine treatment of fever" at the cellular level.Emodin,chrysophanol,cinnamic acid,(+)-catechin,(-)-epicatechin and gallic acid can realize "hot medicine for cold disease" at the cellular level.`... |
