| Background and Purposes:In the process of understanding and curing the disease in traditional Chinese medicine,it is of great significance to clarify the "cold and heat" attribute of the disease for determining the treatment plan."Cold and heat" have different forms in the patient’s physical signs,constitution and traditional Chinese medicine."Treating cold with hot drugs and treating heat with cold drugs" could adjust the "cold and hot" condition of the body with different physical signs and constitutions to restore the normal balance,based on the "cold and hot" bias of traditional Chinese medicine.At present,Chinese oncology has initially formed a complete treatment system.It is often used as the basic treatment for tumor prevention and treatment,focusing on strengthening the foundation and clearing away heat and toxic substances.The pathogenesis of tumor is complex,accompanied by the changeable condition changeable,involving multiple systems and organs,and cold and heat deficiency and excess are mixed.Therefore,in the process of tumor treatment,only by accurately grasping its "cold and hot" attribute,can we accurately use drugs.At present,there is no consensus on the modern biological explanation of "cold and heat",but it suggests that it may be related to Thermo-TRP.So does Thermo-TRP play an important role in the differentiation of "cold and hot" attributes of tumors?The purpose of this project is to explore and verify the expression mode of Thermo-TRP in tumors and the correlation with the molecular mechanism in the anti-tumor process of traditional Chinese Medicine guided by "cold-heat" theory by using a variety of databases,based on the experimental methods of bioanalysis,and combined with the previous and existing research results.Methods and results:In the first part of this subject,based on a variety of databases related to tumor gene expression information and methods of biological information analysis,it is found that at the gene level,Thermo-TRP have different relative expression patterns in different tumors,which has a significant correlation with the prognosis and survival time of patients,so it can be used as the key basis for distinguishing the "cold and hot" attribute of tumors.In the second part of this subject,firstly,based on the preliminary research results in the laboratory,it is verified that the "cold and hot" tumors proposed in the first part conform to the "cold-hot" theory under the intervention of components of traditional Chinese medicine components with "cold and hot" properties.Then the typical ThermoTRP molecules TRPV1 and TRPM8 were used as template molecules to screen potential Thermo-TRP inhibitors in common anti-tumor traditional Chinese medicine ingredients,and then their targets were predicted.It was found that the relative expression of Thermo-TRP had a certain impact on tumor metabolism.In the third part of this subject,based on the tumor differential expression information obtained in the first part,we analyzed the metabolism in different tumors and found that the metabolic genes were relatively down-regulated in "cold" tumors and upregulated in "hot" tumors.Subsequently,taking lung adenocarcinoma and lung squamous cell carcinoma as the representatives of "cold and hot" tumors,it was found that in "cold" tumors,the relatively high expression of TRP sensitive to lower temperature was negatively correlated with metabolic genes,while in "hot" tumors,the relatively high expression of TRP sensitive to higher temperature was positively correlated with metabolic genes.In a word,the relative expression pattern of ThermoTRP in tumors leads to the difference of metabolism between "cold and hot" tumors.Traditional Chinese medicine with obvious "cold and hot" attribute may correct the metabolic abnormalities in tumor patients through Thermo-TRP,so as to achieve the therapeutic effect.Conclusion:Based on different interpretations of "cold and heat" at present,this topic puts forward the conjecture that Thermo-TRP may be the key basis for distinguishing the "cold and hot" attributes of tumors.Based on the combination of bioinformatics analysis methods and various databases,we has found the expression pattern of Thermo-TRP in tumors,and explained the internal mechanism of metabolic differences in tumor patients to a certain extent.It has made a contribution to the accurate differentiation of "cold and heat" of tumor,and has important guiding significance for the establishment of treatment plan for tumor patients in the process of clinical treatment. |
