| In recent years,Ⅱ type diabetes mellitus(T2DM)is one of the high incidence of chronic diseases.Owing to the lack of long-term,reliable and highly compliant treatment methods,T2 DM has become one of the major chronic diseases threatening human health.After the improvement of living conditions,the main reason for the onset of T2 DM is the change of diet and high-calorie foods,and scientific and reasonable regulation of diet has become the main means to prevent and control T2 DM.As a traditional Chinese medicine,Kudzu is rich in natural Kudzu powder,which has high value of both medicine and food.According to the structural characteristics and physical and chemical properties of the resistant starch producted by different methods,the production methods of high anti-digestibility of resistant starch from Kudzu were screened out.On this basis,from the perspective of regulating insulin resistance and intestinal environment,the dose-effect relationship of Kudzu resistant starch in lowering blood glucose was explored through animal experiments,and then the mechanism of the regulation of blood glucose function of Kudzu resistant starch was clarified.(1)Production technology and analysis of physicochemical properties of Kudzu resistant starch.In this study,Kudzu powder was used to prepare autoclaving Kudzu resistant starch,A-KRS,Autoclaving-debranching Kudzu resistant starch,DA-KRS and Purification Kudzu resistant starch,P-KRS.SEM,FT-IR DSC and other analytical methods were used to investigate the structural and physicochemical characteristics of Kudzu resistant starch.In addition,the digestion process of Kudzu resistant starch was simulated by a two-stage in vitro digestion model.The results showed that the three production methods could all make the powder recrystallize,and the treated Kudzu resistant starch particles showed an amorphous block structure,and the surface showed a gully shape with holes.Kudzu resistant starch crystalline region area increases,the thermal stability increased at the same time,compared to the Kudzu powder,three of the four kinds of resistant starch increased the To,Tp and Tc are different,ΔH of P-KRS is significantly enhanced,the P-KRS resistant starch content of 81%,in the digestive power curve reflects the similar results,three kinds of Kudzu resistant starch digestion rate and prediction index were significantly lower blood glucose.(2)Evaluation of the function of Kudzu resistant starch in regulating blood glucose.Dietary intervention was conducted on T2 DM mice induced by high sugar and high fat feed combined with STZ by means of resistant starch from Kudzu,and the effects of different doses of resistant starch from Kudzu root on glucose and lipid metabolism and insulin function in T2 DM mice were studied.At the same time,from the IRS-1/PI3K/Akt/Glut4 signaling pathway,further explore the mechanism of Kudzu resistant starch alleviating insulin resistance.It turned out that the change was 4 weeks later,the mice in PC were abnormally "emasciated" and their body weight decreased by 15%.The body weight of mice in RSH group,RSM group,RSL group and Met group all showed an increasing trend.Four weeks after the diet change,the blood sugar and glucose tolerance of T2 DM mice changed significantly,It is suggested that Kudzu resistant starch can effectively alleviate the abnormal increase of blood glucose in T2 DM mice.Four blood lipid test results show.Four blood lipid test results showed that the intervention of resistant starch from Kudzu could effectively synthesis of insulin,directly affect the storage of liver glycogen,Increase the expend and utilization of glucose by the liver in mice,and significantly reduce the blood sugar content of T2 DM mice..(3)Effects of Kudzu resistant starch on intestinal barrier function and intestinal flora in T2 DM mice.By studying the Kudzu resistant starch to the improvement of T2 DM intestinal barrier function in mice and adjust the function of intestinal flora,lavage after 4 weeks for each colon tissue of mice were analyzed,and the intestines of mice found that T2 DM tight junction protein,higher adhesion protein expression and serum endotoxin content also increased significantly,the tip resistance of Kudzu starch intervention to repair T2 DM intestinal permeability in mice.Meanwhile,Kudzu resistant starch can effectively restore the intestinal homeostasis of T2 DM mice.Compared with PC group,the fecal content of short-chain fatty acids in RSH group,The short-chain fatty acids in the RSM group and RSL group were significantly increased,especially the content of propionic acid and butyric acid was the most obvious,and the increase rate was higher than that in Met group.Kudzu resistant starch can inhibit the further deterioration of intestinal flora disorders in T2 DM mice to a certain extent,and has a better function of regulating intestinal flora in T2 DM mice,Mainly through increasing the abundance of Firmicutes and Bacteroidetes,and significantly inhibiting the proliferation of Proteobacteria and Verrucomicrobia to play its role. |
