| Objective Deep Ⅱ degree scald model rats were used as research objects,research sanguisorba ointment on the role of skin wound healing in rats,and study its mechanism of promoting wound healing.MethodThe 126 female SD rats were selected and randomly divided into normal group,model group,mei-bao’s scald ointment group(MEBO group),mupirocin group,the low-dose group of sanguisorba ointment(0.25g/g),the middle-dose group of sanguisorba ointment(0.5g/g)and the high-dose group of sanguisorba ointment(0.75g/g),each with18 rats.The normal group only had regular hair removal,and the back of rats in the other groups were subjected to high temperature scald method to establish a deep second degree scald model.The burn wound was about 10cm2.After modeling,the drug was administered immediately,wash the wound with normal saline,and then apply the corresponding ointment to the wound with a thickness of about 1.5mm,and the wounds of each group remain open.The normal group and the model group are only given normal saline.Twice a day,with an interval of 12 hours,for 21 consecutive days.On the 7,14,and 21 days,6 rats were randomly selected from each group to observe the general situation of the wounds;the wound healing rate was calculated;the inflammatory factors IL-6 and TNF-α in the serum of rats were detected by ELISA;HE staining method to observe the pathological changes of wound tissue;immunohistochemical method to detect the expression of CD31 protein in wound tissue;Western blot method to detect the relative expression content of VEGF protein in wound tissue.Results1.Gross morphological observation showed that the wounds were observed daily after modeling.The burned parts of rats in each group were grayish white after modeling,and the skin elasticity decreased;after scalds,the wounds of rats in each group first showed swelling,exudation,granulation tissue formation,and then gradually formed crusts.Among them,the high-dose of sanguisorba ointment group healed fastest,the mei-bao’s scald ointment group and the medium-dose of sanguisorba ointment group followed,and the low-dose of sanguisorba ointment group and the mupirocin group were poor.2.Wound healing rate measurement results showed that on the 7,14,and 21 days after scald,the wound healing rate of mei-bao’s scald ointment group and the high and medium dose of sanguisorba ointment groups were significantly higher than that of model group(P<0.05),indicating that the high and medium dose of sanguisorba ointment and mei-bao’s scald ointment has a good effect on promoting wound healing.Compared with mei-bao’s scald ointment group,there was no significant difference in wound healing rate of high-dose of sanguisorba ointment group on the 7th and 21 st days after scald(P>0.05),and the wound healing rate of high-dose of sanguisorba ointment group was significantly increased on the 14 th days after scald(P<0.05),indicating that the high-dose of sanguisorba ointmen is slightly better than mei-bao’s scald ointment in promoting wound healing.On the 7,14,and 21 days after scald,there was no significant difference in wound healing rate in the middle-dose of sanguisorba ointment group and mei-bao’s scald ointment group(P>0.05),mupirocin group and low-dose of sanguisorba ointment group were also(P>0.05),indicating that the medium-dose of sanguisorba ointment and mei-bao’s scald ointment have the same effect on promoting wound healing,and low-dose of sanguisorba ointment and mupirocin have similar healing effects.On the 7th and 14 th day after scald,the wound healing rate of low-dose of sanguisorba ointment and mupirocin group was significantly lower than that of mei-bao’s scald ointment group(P<0.05),indicating that the healing effect of low-dose of sanguisorba ointment and mupirocin was lower than that of mei-bao’s scald ointment group.Experimental data showed that the high-dose of sanguisorba ointment group had the fastest healing speed,followed by the medium-dose of sanguisorba ointment group and mei-bao’s scald ointment group,and the low-dose sanguisorba ointment group and the mupirocin group were the last.3.Determination of IL-6 and TNF-α in serum showed that on the 7,14,and 21 days after scald,the serum levels of inflammatory factors IL-6 and TNF-α in the model group were higher than normal group(P<0.05);the levels of inflammatory factors gradually decreased over time,and the change trend of each administration group is the same as the model group.On the 14 th and 21 st days after scald,the levels of inflammatory factors IL-6 and TNF-α in the high-dose of sanguisorba ointment group were significantly lower than those in the mupirocin group(P<0.05),indicating that the high-dose of sanguisorba ointment had better antibacterial effects than mupirocin.At 7,14,and 21 days after scald,the levels of inflammatory factors IL-6 and TNF-α in the medium-dose of sanguisorba ointment group were not significantly different from those in the mupirocin group(P>0.05),suggesting that the medium-dose of sanguisorba ointment has similar antibacterial effects with mupirocin.On the 7th day after scald,the levels of inflammatory factors IL-6 and TNF-α in the mupirocin group were significantly lower than those in the mei-bao’s scald ointment group(P<0.05),indicating that the antibacterial effect of mupirocin was better than mei-bao’s scald ointment.On the 14 th and 21 st days after the scald,the IL-6 level of the mei-bao’s scald ointment group was significantly lower than that of the low-dose of sanguisorba ointment group(P<0.05),on the 7th and 21 st days after the burn,the level of inflammatory factor TNF-α in mei-bao’s scald ointment group was significantly lower than that of low-dose of sanguisorba ointment group(P<0.05),indicating that the antibacterial effect of mei-bao’s scald ointment was better than the low-dose of sanguisorba ointment.It showed that the high-dose of sanguisorba ointment had good antibacterial effect,followed by medium-dose of sanguisorba ointment and mupirocin,mei-bao’s scald ointment group and low-dose of sanguisorba ointment were not good.4.Histopathological results of wound showed that in the normal group,the structure of each layer was clear and complete,the hair follicles and sebaceous glands were evenly distributed,and the dermal layer tissue and accessory structure were clear.On the 7,14,and 21 days after scald,compared with the model group,the formation of new capillaries and sweat glands in the epidermis and dermis in the high-dose sanguisorba ointment group and mei-bao’s scald ointment group were significantly faster,and the healing effect were the best;The formation speed of fibroblasts,new capillaries and collagen fibers in the mupirocin group and the low and medium dose groups of sanguisorba ointment were similar.5.Immunohistochemistry results showed that rats in the normal group and each scald group expressed CD31 at all time points after scald,and reached the peak on the 14 th day.On the 7,14,and 21 days after scald,the relative expression of CD31 on the wound of the model group was higher than normal group(P<0.05),and the relative expression of CD31 on the wound of the high and medium doses of sanguisorba ointment groups and mei-bao’s scald ointment group were significantly higher than that of the normal group(P<0.05).On the 14 th and 21 st days after the scald,the relative expression of CD31 in the low-dose of sanguisorba ointment group and the mupirocin group was significantly lower than that of the mei-bao’s scald ointment group(P<0.05),and there was no significant difference in the relative expression of CD31 on wounds in the high and medium-doses of sanguisorba ointment groups and mei-bao’s scald ointment group at 7,14,and 21 days after the scald(P>0.05).It showed that the high and medium doses of sanguisorba ointment and mei-bao’s scald ointment had a better effect of promoting the formation of wound microvessels,followed by low-dose of sanguisorba ointment and mupirocin.6.Western blot result showed that at 7,14,and 21 days after scald,the expression of VEGF protein in the wound of the model group was higher than that in the normal group(P<0.05);the expression of VEGF protein in the high and medium doses of sanguisorba ointment groups,mei-bao’s scald ointment group and mupirocin group were significantly higher than that of the model group(P<0.05).On the 14 th and 21 st days after the scald,the expression of VEGF protein in the high and medium doses of sanguisorba ointment groups were not significantly different than that of mei-bao’s scald ointment group(P>0.05),indicating that high and medium dose of sanguisorba ointment and mei-bao’s scald ointment group has a similar effect on promoting angiogenesis effect.ConclusionSanguisorba ointment can obviously promote the deep Ⅱ degree burn wound healing in rats,promote the formation of new blood vessels,inhibit wound infection,improve the quality of healing.It may be related to reducing the production of inflammatory factors IL-6 and TNF-α,increasing the synthesis and release of CD31 and VEGF in scald wound tissue,and promoting wound microangiogenesis. |
