The Influence Of ShengJi BaiYu Ointment On The Expression Of BFGF And TGF-β1 In The Deep Ⅱ Degree Burn Wound In Rats

Objective: Preliminary study of the effects of shengjibaiyu ointment on the distribution and quantities of rats with the deep II degree burn wound of b FGF and TGF-β1, and its relationship with wound healing between shengjibaiyu ointment, further understanding the mechanism of promoting burn wound healing.Methods: 128 Healthy SD(SPF) were chosen, male and female for each half, body weight(200 ± 20)g, after a week adaptive feeding with randomly divided into blank group, model group, control group, experimental group, each group were 32 rats. Before the burn day, using depilatory creams, area of about 4cm in diameter, water wash, wipe. Before the experiment rats fasting in 12 hours, then freely drinking. Except the blank group, the other three groups with 10% chloral hydrate intraperitoneal injection of 300 mg/kg in anesthetized rats, immersed in the back of of constant temperature of 100 ℃ water bath for 5 seconds, causing deep second degree burn(confirmed by pathology). The experimental group after modeling shengjibaiyu ointment topical wound immediately 1g; the control group with external application of MEBO 1g; model group outside the rub saline; the 3 groups were daily change medicine 1 times, continuous administration to put to death 1 days ago. Do not give any treatment of blank group, normal feeding. The above four groups use the medicine two times a day, a total of two weeks of treatment. Normal control group did not give any treatment. In the model after 3 d, 7 d, 14 d, and 21 d, to observe the record each wound healing, using image analysis software for measuring each wound area, and the wound granulation tissue production of tissue sections, HE staining pathological observation, use SABC immunohistochemical method for the determination of components in granulation tissue of b FGF, TGF-β1 content, and statistical analysis of data.Results: After 3, 7, 14 and 21 d, wound areas of experimental rats were significantly reduced. At the same time, the wound healing rate of shengjibaiyu ointment group and MEBO group showed significantly higher than the model group with statistical difference(P<0.01), but shengjibaiyu ointment group and MEBO group showed no significant difference(P>0.05). After 3 d, the wounds of 4 experiment groups were main performance of seepage and hyperemia, and relatively rare. After 7 d, the model group did not see any clear capillaries as well as epithelial hyperplasia. Born muscle jade cream and Mabel moist burn cream group see lots of blood capillary hyperplasia and proliferation of fibroblasts. A small amount of inflammatory cells infiltration were showed, began to form, collagen hyperplasia of epithelial cells saw a small amount to show. The two groups of Shengjibaiyu ointment and MEBO showed a large number of capillaries and fibroblasts hyperplasia, but the epithelial hyperplasia were not obvious to show. After 14 and 21 d, rats in model group could see a small amount of formation of granulation tissue, a rare fiber cells, epithelial hyperplasia, collagen fibers and irregular arrangement. But the groups of Shengjibaiyu ointment and MEBO showed more formation of granulation tissue, collagen formation, obviously longitudinal arrangement, epithelial cells, and fibroblasts hyperplasia.After 3 d, b FGF expression difference in the experimental rats of 4 groups were not big, and had a higher degree of positive expression(++). After 7 d, the model group rats began to decline, positive expression of b FGF quantity(+), and in Shengjibaiyu ointment group and MEBO group increased(+++). After 14 and 21 d of modeling, the positive expression of TGF-β1 in granulation tissue of skin wounds of rats in 4 groups showed a declining trend, the expression in model group showed(- ~+), but in Shengjibaiyu ointment group and MEBO group showed no significant difference(P > 0.05).After 3 d,the expression of TGF-β1 of skin wound granulation tissue of 4 groups experimental rats had no significant difference, but showed higher degree of positive expression(++). After 7 d, the expression of TGF-β1 of skin wound granulation tissue of model group began to decline and showed(+), but Shengjibaiyu ointment group and MEBO group increased and showed(+++), and the two groups were significantly higher than that of model group. After 14 and 21 d, positive expression of of TGF-β1 in experimental rats of four groups showed declining trend; model group showed(- ~ +), but Shengjibaiyu ointment group and MEBO group showed(+),and still higher than that of model group. Shengjibaiyu ointment group and MEBO group showed significant differences(P < 0.01) compered with model group,but Shengjibaiyu ointment group and MEBO group showed no significant difference(P > 0.05).Conclusion:(1) Shengjibaiyu ointment can obviously promote the wound’s healing, improve the healing rate and healing qualities, and basically showed the same effects with MEBO;(2) The mechanism of promoting wound healing of Shengjibaiyu ointment should be related with the modulation of cutaneous wound granulation tissue in the expression of b FGF and TGF-β1.
