| Objective:To study the expression and clinical significance of autophagy related protein antitubule related protein 1(microtubule-associated protein 1light chain 3,LC3)and Beclin-1 in peripheral blood of children with mycoplasma pneumoniae pneumonia(MPP).Methods:Collected the sample blood of mild and severe mycoplasma pneumoniae pneumonia(MPP)patients hospitalized in Shanxi Provincial people’s Hospital from January2018 to May 2020 and the peripheral blood of children with normal outpatient examination.Next we separated the white blood cells,detected the LC3II、LC3I、Beclin-1 expression in white blood cells by using Western Blot and calculated the LC3II/I ratio;Next,we detected the content of interleukin-6(IL-6)、tumor necrosis factor-a(TNF-α)with ELISA.Compared the expression of the indexes above in serum of three groups of children with mycoplasma pneumoniae pneumonia and analyzed the correlation between autophagy and cytokines.Results:A total of 50 cases were included in the MPP group,including 28 cases of mild disease,22 cases of severe disease and 25 cases of control group.1.The ratio of autophagy protein LC3 II to LC3 I and Beclin-1 expression in peripheral leukocytes of MP mild group and severe group were higher than those control group.MP severe group was higher than the mild group.The difference was statistically significant(P<0.05).2.MPP the IL-6、TNF-α level of mild and severe group were higher than those of control group.The IL-6、TNF-α level in MPP severe group were higher than those in mild group.The difference was statistically significant(P<0.05).3.The ratio of autophagy protein LC3 II to LC3 I and Beclin-1 and IL-6、TNF-α were positively correlated(P<0.05).Conclusion:1.Autophagy occurs in peripheral leukocytes in MPP,autophagy is related to the degree of illness,the more serious the disease,the higher the degree of autophagy.2.The inflammatory response is obvious when Cytokines increases in peripheral leukocytes in MPP.3.Autophagy was positively correlated with cytokines,suggesting that autophagy in the peripheral blood leukocytes of MPP children affected the change of cytokines,thus affecting the severity of the disease. |
