Therapeutic Rules And Core Drug Network Pharmacological Analysis Of Interstitial Lung Disease By Prof.zhixiu Yang

Abstract/Summary:
Objective:To explore the medication rule of Professor Yang Zhixiu in the treatment of interstitial lung disease(ILD),and to study the possible mechanism of his core drug prescription in the treatment of idiopathic pulmonary fibrosis(IPF).Method:All the patients who met the diagnosis of interstitial lung disease in the outpatient department of Professor Yang Zhixiu,Nangang Branch of Heilongjiang Provincial Hospital of traditional Chinese medicine from March 2019 to March 2021 were collected.After processing the prescription information of patients,it was imported into SPSS statistics 26 and SPSS modeler 18 for cluster analysis and association rule analysis,and visualized with Cytoscape 3.8.0 to explore the medication rules and core prescription drugs of Professor Yang Zhixiu in the treatment of ILD.Then tcmsp database was used to query the effective ingredients and target information of core drug prescriptions.At genecards(https://www.genecards.org/)And drugbank(https://go.drugbank.com/)The two databases were searched for IPF related targets,and the targets in the two databases were integrated to get the disease targets of IPF.The core drug targets were matched with IPF disease targets to obtain coincidence targets,and then string was used to construct protein-protein interaction(PPI)network.PPI network information is imported into cyberscape 3.8 for further analysis,and core targets are screened.The gene ontology(go)enrichment analysis and Kyoto Encyclopedia of genes and genomes(KEGG)signal pathway were used to analyze the core target using metascape database,and the potential mechanism of the prescription treatment of IPF was speculated.Result:(1)Among the 256 prescriptions selected,201 drugs were used,and the top 10 drugs used most frequently were: fried licorice,Trichosanthes,forsythia,honeysuckle,tangerine peel,Fritillaria thunbergii,bitter almond,ginkgo,cortex mori and semen Raphani.The order of the four Qi usage was cold,warm,flat,cool and hot.The order of the five flavors usage was bitter,sweet,pungent,salty,astringent,sour and light.The order of the meridian tropism usage was lung,stomach,spleen,heart,liver,kidney,large intestine,small intestine,bladder,gallbladder,Sanjiao and pericardium.Through data mining,we found that there were six commonly used drug pairs,honeysuckle and forsythia;Aster,Tussilago farfara;Trichosanthes kirilowii and cortex mori;Radix Glehniae,Radix Ophiopogonis;Tangerine peel,Pinellia ternate;Stir fried yam and dangshen with bran.Through cluster analysis,we can get the prescription rules of Yang Shi’s treatment of ILD,and through association rules,we can find that Forsythia suspensa,Fritillaria thunbergii,honeysuckle,Trichosanthes kirilowii,fried licorice and almond are the basic prescriptions of Yang Shi’s treatment of ILD.Through the complex network analysis,the core drug prescriptions were processed licorice,Trichosanthes,forsythia,honeysuckle,tangerine peel and Fritillaria thunbergii.(2)Based on the network pharmacology method,the possibility of IPF treatment with core drug prescription was analyzed.The main effective components of core drug prescription were naringenin,wogonin,beta sitosterol,kaempferol,luteolin and quercetin,and the core targets were AKT1、TP53、IL6、MAPK1、VEGFA、TNF.Go analysis showed that the treatment of IPF with core drug prescription may be related to the regulation of DNA binding transcription factor activity,response to growth factors,vascular development,inflammatory reaction,epithelial cell proliferation,capsule cavity,membrane raft,RNA polymerase II transcription regulatory complex,cytoplasmic perinuclear region,ficolin-1rich granular cavity,and other biological processes Endoplasmic reticulum and other cellular components may play an important role in the treatment of IPF.Cytokine receptor binding,kinase binding,transcription factor binding,nuclear receptor activity,phosphatase binding,core promoter sequence specific DNA binding and other molecular functions may play an important role in the treatment of IPF.Through KEGG enrichment analysis,it is found that the core drug prescription treatment of IPF may be related to the pathway of cancer Epstein Barr virus infection,lifespan regulation pathway,p53 signaling pathway,TGF-β It is related to signal pathway,AMPK(adenylate activated protein kinase)signal pathway,antigen processing and presentation,calcium signal and so on.Conclusion:Through data mining technology,it is found that Professor Yang Zhixiu’s outpatient treatment for ILD patients is mainly to remove pathogenic factors,and also to strengthen the body.He mainly uses heat clearing,detoxification,expectorant,antitussive and antiasthmatic drugs to remove the evil Qi in the lung,reduce the adverse Qi in the lung,and supplement the products of Supplementing Qi and nourishing yin to help healthy qi and evil go out.Finally,he uses the drugs of regulating qi and dredging collaterals to remove the obstruction in the lung.The core prescriptions were Radix Glycyrrhizae,Fructus Trichosanthis,Fructus Forsythiae,honeysuckle,Pericarpium Citri Reticulatae and Fritillaria thunbergii.The main effective components of the core prescription were
