BackgroundPsoriasis is a common chronic inflammatory skin disease.The pathogenesis of psoriasis has not yet been fully elucidated,but the involvement of immune cells and immune molecules in the occurrence and development of psoriasis has been confirmed.IL-33 is the member of IL-1 cytokine family,and ST2 is specific ligand of IL-33.Numerous studies have demonstrated that IL-33 and ST2 increased in lesions of psoriasis vulagaris,but the correlation between their levels in serum and psoriasis severity is still controversial.Therefore,this study aims to explore the levels of IL-33,ST2,IL-17 and IL-5 in peripheral blood of PsV patients and their clinical significance,so as to provide a basis for further understanding the pathogenesis of PsV and searching for therapeutic targets.ObjectiveThe objective of this study was to detect the levels of IL-33,ST2,IL-17 and IL-5 in the peripheral blood of PsV patients,analyze the correlation between their expressions and PASI(Psoriasis area and severity index)scores,so as to investigate their role and significance in clinical diagnosis and treatment of PsV patients.MethodsA total of 68 patients with PsV diagnosed clinically in the department of Dermatology and Venereal diseases in the First Affiliated Hospital of Xinxiang Medical University between September 2018 to April 2019,including 43 patients in the active stage,11 in the stationary stage,and 14 in the retrograde stage.Sixty healthy volunteers were collected from the Health Examination Center of the First Affiliated Hospital of Xinxiang Medical University between December 2018 to April 2019.Peripheral venous blood of PsV patients and healthy control group was collected on an fasting basis in the morning.Serum were separated by centrifugation and serum expression levels of IL-33,ST2,IL-17 and IL-5 were measured by enzyme-linked immunosorbent assay(ELISA).Statistical analysis of the data was performed by using Statistical Package for Social Sciences(SPSS)soft,version 22.0.If the data satisfied normal distribution,data were expressed as mean±standard deviation(Mean±SD),Student’s t test and One-way ANOVA were used to analyze different groups,Pearson was used for correlation analysis;otherwise data were expressed as M(P25,P75),Mann-Whitney U test was used for comparative analysis,Kruskal-Wallis H test was used for comparison among multiple groups,Spearman was used for correlation analysis.A P-value≤0.05 was defined statistically significant.Results1.Compared with the healthy control group,the serum levels of IL-33,ST2,IL-17and IL-5 in PsV patients were significantly increased(P<0.05).2.Compared with the healthy control group,serum IL-33,ST2,IL-17 and IL-5levels in active stages were higher than those in controls(P<0.05);serum ST2 and IL-17levels in active stages were higher than those in retrograde stages(P<0.05).There was no significant difference between other groups(P>0.05).3.Serum IL-33 and IL-17 expression levels in PsV patients were positively correlated with PASI scores(P<0.05).Conclusion1.The serum levels of cytokines IL-33,ST2,IL-17 and IL-5 were significantly increased in patients with PsV,especially during the active stages,suggesting that IL-33,ST2,IL-17 and IL-5 are involved in the pathological mechanism of PsV,which can reflect the disease activity of patients with PsV.2.The levels of IL-33 and IL-17 in PsV patients were positively correlated with PASI score,indicating that compared with IL-5 and ST2,the expression levels of IL-33and IL-17 may be more representative of the severity of PsV.
