| Objective:To observe the mutation of IL-23R gene polymorphism in the population of the Ningxia region.To analyze polymorphisms of Interleukin-23 Receptor(IL-23R)and susceptibility to Psoriasis Vulgaris(PV)from Ningxia region.Methods:All of the participants were come from NingXia region.There were 101patients with psoriasis vulgaris(PV)and 103 normal healthy controls in the study.Six SNPs(rs10489629、rs1343151、rs10889677、rs11209032、rs1495965、rs1004819)of IL-23R gene were genetyped by using the Taqman fluorescent PCR.The results were verified by Sanger Gene sequencing.All analyses were performed with SPSS 21.0 and Haploview 4.2software.Comparisons between the two groups were performed with Hardy-Weinberg equilibriumChi-square test and hyplotype analysis.Results:1.The SNP sites of the IL-23R gene(rs10489629、rs1343151、rs10889677、rs11209032、rs1495965、rs1004819)were consistent with the Hardy-Weinberg equilibrium.2.There were no significant difference in the genotype and allele frequency of IL-23R gene’s SNP rs10489629、rs1343151、rs10889677、rs11209032、rs1495965、rs1004819 between PV patients and normal controls(P>0.05).3.There were also no significant difference between Hui people and Han people in the PV patients.4.There was a linkage disequilibrium between rs11209032 and rs1495965 SNPs(D’=0.957,r~2=0.821)by Linkage Disequilibrium(LD)test.Further analysis revealed there was no significant difference in haplotype of rs11209032 and rs1495965(P>0.05).5.The Sanger sequencing result was as same as the genetyed by the Taqman fluorescent PCR.Conclusion:The susceptibility to psoriasis vulgaris(PV)may not to be associated with the SNPs rs10489629、rs1343151、rs10889677、rs11209032、rs1495965 and rs1004819 of IL-23R gene in Ningxia region. |
