| Objective:TH17 is a new discovered CD4~+T,which is different from TH1,H2.It does not produce IL-4 or IFN-γ,but specially release IL-17A(IL-17),IL-17F,IL-22. TH17 has independent mechanism of differentiation and developmental regulation.It plays important role in autoimmune disease and infectious disease,so it becomes a hot study recently.Therefore this study is to investigate the corelationship between TH17 related cytokines IL-23,IL-17A,IL-17F,IL-22 and psoriasis vulgaris or vitiligo,and further to illustrate the pathogenesy of psoriasis vulgaris and vitiligo.Methods:According to the selected standard,we collected 30 patients of psoriasis vulgaris.15 patients in active phase,and 15 patients in stable phase.And selected 30 patients of vitiligo,with 15 patients in active phase,and 15 patients in stable phase. And collect 30 healthy controls.Then we drawed peripheral blood and to do the next experiments.1.The level of IL-23 in plasma was detected by ELISA,and analyzed the differernce between active or stable psoriasis and the healthy controls.The level mRNA expression of IL-17A,IL-17F and IL-22 in peripheral blood mononuclear cell (PBMC) were also detected by SYBR Green I real-time RT-PCR,and analyzed the differernce between active or stable psoriasis and the healthy control.2.Use the same methods to analyze the differernce between the vitiligo in active or stable stage and the healthy controls.Results:1.The level of IL-23 in active psoriasis was significantly higher than that of healthy control group(P =0.002).But The level of IL-23 in stable psoriasis was not significantly higher than that of the control group(P =0.169).There was no difference between in active psoriasis and in stable psoriasis(P =0.192).The level of IL-17A mRNA,IL-17F mRNA,IL-22 mRNA in active psoriasis were significantly higher than that of the control group respctively(P =0.007;P =0.015;P =0.002).But no significance has been observed between the stable psoriasis and the control(P =0.8755;P =0.675;P =0.55).The level of IL-17A mRNA,IL-22 mRNA but IL-17F in active psoriasis was significantly higher than that in stable psoriasis. 2.The level of IL-23 in active and stable vitiligo was significantly higher than that of the control group(P =0.01,P =0.033) ).The level of IL-23 showed no differernce between active and stable vitiligo(P =0.972).The level of IL-17A mRNA in active vitiligo was significantly higher than that in stable vitiligo and the control group(P =0.016;P =0.001).But it had no significance between the stable vitiligo and the control group(P =0.991).The level of IL-17F mRNA,IL-22 mRNA in active or stable vitiligo was not higher than that of the control group significantly.Conclusion:1.The level of IL-23 in plasma,the level of IL-17A mRNA,IL-17F mRNA, IL-22 mRNA in active psoriasis were significantly higher than that of the control group.These demonstrated that TH17 related cytokines IL-23,IL-17A,IL-17F, IL-22 participated in psoriasis vulgaris.2.The level of IL-23 in active and stable vitiligo and the level of IL-17A mRNA in active vitiligo were significantly higher than that of the control group. These indicated that IL-23,IL-17A participate in vitiligo,but we need more study to make sure the corelationship between IL-17F,IL-22 and vitiligo.3.There may be a new therapy to treat psoriasis vulgaris by blocking IL-23/TH17 or TH17 related cytokines IL-23p40,IL-17,IL-22.But we need more research to make sure that it has a relationship between IL-23/TH17 and vitiligo. |
