Objective:To explore the experimental study on the prevention and treatment of intrauterine adhesions with the Tibetan medicine weisenia pill.By observing the morphology of uterus and endometrium and comparing the degree of fibrosis,we explored the role of weisenia pill in the prevention and treatment of intrauterine adhesions.By observing the expression levels of TGF-β and MMP-9,we studied the formation mechanism of intrauterine adhesions at the molecular level,so as to provide theoretical basis for the prevention and treatment of intrauterine adhesions The experimental basis of medicine.Methods : Thirty-five 6-8-week-old female Sprague Dawley rats weighing180-200 g were selected.All animals were reared adaptively for one week,drinking and feeding freely under temperature [(23 ± 2)℃] and light(light 12 h and dark 12h).In the first part of the experiment,15 rats were randomly divided into three groups:normal group,model I group and study I group,in which the normal group was not treated with any treatment,model I group and study I group were made by mechanical injury and alcohol burning method,and study I group was given the Tibetan medicine weisenia pill by gavage immediately after modeling for 14 days;in the second part,10 rats were randomly divided into two groups In the third part,10 rats were randomly divided into two groups: model group III and study group III.mechanical injury and alcohol burning were also used in the two groups.The rats in study group III were given weisenia pill for 14 days from the 10 th day after modeling Weisenia pill was given orally for 14 days.In each part of the experiment,the uterine tissues of all rats were taken out 14 days after treatment,and the morphology of all uterus was observed.The tissues were fixed in 4% formaldehyde and made into paraffin sections24 hours later.He staining was used to compare the morphology of endometrium and the number of endometrial glands;Masson staining was used to compare the area of endometrial fibrosis;immunohistochemical staining was used to compare the effect of weisenia Pill on the expression of TGF-β and MMP-9.Results :(1)Compared with the normal group(19.9681 ± 0.3254)μm,the endometrial thickness of study group I(17.7648 ± 0.5796)μm had no significant difference(P > 0.05),but compared with the model group I(13.3920 ± 0.2044)μm,the endometrial thickness of study group I gradually increased(P < 0.05);study group II(16.1066 ± 0.2607)The endometrial thickness of model group Ⅱ was(14.3677 ±0.4997)μm,but the endometrial thickness of model group Ⅱ was(14.3677 ± 0.4997)μm,The thickness of endometrium in study group III(15.4876 ± 0.4060)μm was not significantly thicker than that in model group III(14.6215 ± 0.0538)μm(P > 0.05),but was thinner than that in normal group(P < 0.05).(2)There was no significant difference in the number of endometrial glands between study group I(38.2267 ± 2.8674)and normal group(44.2220 ± 3.0912),but gradually increased compared with model group I(25.3030 ± 1.2472)(P < 0.05);there was no significant difference in the number of endometrial glands between study group II(27.2568 ± 2.0548)and model group II(23.3004 ± 1.2472)(P > 0.05),There was no significant difference in the number of endometrial glands between study group III(29.1941 ± 2.8674)and model group III(23.6333 ± 1.2472)(P > 0.05),but there was also a decrease compared with normal group(P < 0.05).(3)The percentage of endometrial fibrosis area in study group I was(20.3398 ±3.5013)%,which was not significantly different from that in normal group(14.3573 ±0.1823)%,but decreased compared with that in model group I(53.2503 ±0.4229)%(P<0.05);the percentage of endometrial fibrosis area in study group II was(42.6873±1.7057)%Compared with the normal group,it increased significantly(P<0.01),decreased significantly(P<0.05)compared with the model group II(56.3773±1.9195)%;the percentage of endometrial fibrosis area in study group III(44.6620 ±1.2219)%,increased significantly(P < 0.01),decreased significantly(P < 0.05)compared with the model group III(57.3857 ± 2.0548)%.(4)The expression level of TGF-β in endometrium of study group I(1.1144±0.0619)was lower than that of model group I(3.6498 ± 0.2866)(P < 0.05),but there was no significant difference compared with normal group(0.9795 ±0.0696);the expression level of TGF-β in endometrium of study group II(1.6800 ±0.0906)was higher than that of normal group(P < 0.05),but there was no significant difference compared with model group II(2.3290 ± 0.2812)The expression level of TGF-β in endometrium of study group Ⅲ(1.7953 ± 0.0630)was also higher than that of normal group(P < 0.05),but there was no difference compared with model group Ⅲ(2.2323 ± 0.2075).(5)The expression level of MMP-9 in endometrium of study group I(2.3810 ±0.2000)was higher than that of model group I(0.6500 ± 0.0838)(P < 0.05),and there was no significant difference compared with normal group(2.6938 ± 0.3435);the expression level of MMP-9 in endometrium of study group II(1.3118 ± 0.0548)was higher than that of model group II(0.9620 ± 0.0819),but there was no significant difference between study group II and model group The expression level of MMP-9 in endometrium of rats in group II was lower than that in normal group(P < 0.05);the expression level of MMP-9 in endometrium of rats in study group III(1.4870 ±0.2797)was not significantly different from that in model group III(0.9620 ± 0.0819),but the expression level of MMP-9 in endometrium of rats in study group III and model group III was lower than that in normal group(P < 0.05).Conclusion: The Tibetan medicine weisenia pill has a certain role in preventing the formation of intrauterine adhesions.It can promote the recovery of damaged endometrium and reduce the degree of fibrosis.Its mechanism may be through inhibiting the expression level of TGF-β and up regulating the expression level of MMP-9 to play an anti fibrosis role,so as to prevent the formation of IUA in rats.With the prolongation of modeling time,after the formation of IUA,the application of Tibetan medicine weisenia pill under the same medication conditions could not effectively improve the morphology of endometrium,but had poor effect in regulating the expression levels of TGF-β and MMP-9.
