Based on the theory of traditional Chinese medicine,this paper takes mulberry leaf and silkworm excrement as the starting point of the relationship between food and metabolites,takes the mechanism of component transformation as the core,and carries out in-depth research through modern analytical techniques,so as to provide basis for clarifying the theory of Chinese medicine.The main research contents are divided into the following five chapters:In the first chapter,the chemical components and pharmacological actions of mulberry leaves and silkworm excrement were reviewed.The reported components of mulberry leaves and silkworm excrement contain flavonoids,alkaloids,terpenes,phenylpropanoids and other active components,as well as many pharmacological actions such as hypoglycemic,antitumor,anti-inflammatory,bacteriostatic,antioxidant,and hypolipidemic.Compared with mulberry leaves,the reported silkworm excrement excrement fewer compounds in species and quantity.The second chapter is based on UPLC-Q-TOF-MS technology to analyze the chemical components of mulberry leaves and silkworm excrement.Based on the study and summary of the fragmentation law of the chemical components of mulberry leaves and silkworm excrement by mass spectrometry,the composition analysis of mulberry leaves and silkworm excrement was carried out.90 compounds were obtained from mulberry leaves,24 of which were reported in mulberry leaves for the first time.There are 50 compounds resolved in silkworm excrement,40 of them are reported in silkworm excrement for the first time.In the third chapter,the chemical components of mulberry leaves and silkworm excrement were analyzed.Based on the analysis of chemical components of mulberry leaves and silkworm excrement,the main difference components are flavonoids,chlorogenic acids,fatty acids and nitrogen-containing compounds.Based on the analysis of the main difference components,a method for the determination of 19 chemical components in mulberry leaves and silkworm excrement was established by using UPLC-Q-TRAP-MS-MS technology,and the content differences of various compounds in mulberry leaves and silkworm excrement were observed.The results showed that the content of flavonoids and chlorogenic acids in mulberry leaves and silkworm excrement had the most obvious changes.There are also differences between fatty acids and nitrogenous compounds.The fourth chapter is the in vitro degradation of chemical components of silkworm.In the second and third chapter,it is found that there are differences in chemical composition between fresh and commercial silkworm excrement,which may be caused by the further metabolism of components in the storage of fresh silkworm excrement.Therefore,the differences between fresh and commercial silkworm excrement were analyzed,and the main differences were flavonoids.Fresh silkworm excrement were collected and stored at different time points,and the relative content of different compounds was observed.The fifth chapter the metabolism of rutin and chlorogenic acid in vitro was studied by using crude protease from midgut of silkworm.Based on the analysis of the main difference components of mulberry leaves and silkworm excrement,namely,flavonoids and chlorogenic acid,rutin and chlorogenic acid were selected as the substrates to observe the metabolism of crude protease in the midgut of silkworm.The results showed that the metabolism of chlorogenic acid was obvious and complete within 4 hours,while rutin did not.Through the above research,it is clear that flavonoids and chlorogenic acids are the different components in mulberry leaves and silkworm excrement.At the same time,it is proved by experiments that the metabolism of chlorogenic acid by the crude protease in the midgut of silkworm is obvious,which is the main mechanism of its transformation.Flavonoids and chlorogenic acids are widely existing compounds in plants,which have a variety of biological activities and pharmacological effects.The change of these two compounds may be the main reason for the transformation of mulberry leaves from xin liang xuan san to gan wen hua shi,which provides evidence for the possible mechanism of the transformation of mulberry leaves and components of silkworm excrement.Due to the complexity of the components in traditional Chinese medicine and the different links involved in the process chain from mulberry leaves to silkworm excrement,the transformation of its components needs to be further studied.
