Background:Lung cancer already ranked the first in incidence and mortality among malignant tumors.Common diagnostic methods in clinical practice,such as spiral CT,PET-CT,serum protein tumor markers,pathological biopsy,cannot simultaneously achieve high sensitivity,high specificity,and minimally invasive treatment.Tumor educated platelets(TEPs)are considered a promising liquid biopsy material.It has shown promising value in the early diagnosis and differential diagnosis of lung cancer.It will be of great value If it works in Efficacy monitoring of lung cancer too.purpose:Use next generation sequencing technology and bioinformatics analysis to compare the expression differences of platelet mRNA from venous blood in patients with lung cancer before and after treatment;explore its potential application value as a biomarker in monitoring the efficacy of lung cancer treatment;find the potential combination of TEPs mRNA that may be useful in follow-up Experimental evaluations.Methods:Collect 10 patients with non-small cell lung cancer before treatment,collect 5 patients with non-small cell lung cancer after surgery and postoperative adjuvant chemotherapy to achieve imaging-level cure.use next generation sequencing to obtain the platelet mRNA expression of each sample,and use statistics methods to analysis differential expression of mRNA.Put differentially expressed genes into cluster analysis and functional annotation,selected some genes with the largest differential expression and the best uniformity,and combined these genes with Gene bank,Pubmed and VanFang Data to verify their function.Results:By comparing the results of platelet mRNA sequencing between the pre-treatment and post-treatment groups of lung cancer,315 differentially expressed genes were found,of which 215 were up-regulated in the post-treatment group and 100 were down-regulated in the post-treatment group.Gene Ontology functional analysis suggests that differentially expressed genes are mainly related to signal transduction,immune system process,and cell adhesion in biological processes,Cell components are mainly related to cell membrane,Cytoplasm and nucleus are related,and their molecular functions are mainly related to protein binding,metal ion binding,and transterase activity.Enrichment analysis of the KEGG pathway revealed that the differentially expressed genes are mainly related to Choline metabolism in cancer,EGFR tyrosine kinase inhibitor resistance,Regulation of actin cytoskeleton and transcriptional misregulation in cancer,etc.19 genes with the largest differential expression and the best uniformity,and 9 genes were selected because they are related to lung cancer.and the change trend of these genes in this study was consistent with the change trend in other previous studies.Conclusions:There are significant differences in the gene expression profile of mRNA in the platelets of patients before and after lung cancer treatment,and most of these differentially expressed genes are related to the occurrence and development of tumors.The literature shows that some genes are involved in the occurrence and development of lung cancer,and the trend of change is consistent with the existing studies.It suggested that the tumor educated platelet RNA profile changes before and after lung cancer treatment.It further reveals the potential application value of tumor education platelets in the evaluation of the therapeutic effect of lung cancer.
