Background: Platelets are central factors in both systemic and local responses during cancer growth.Platelets affect cancer cells by transferring cancer-related biomolecules,which lead to the enrichment of these cancer-related biomolecules in platelets,including RNA.Based on this,this study used the next generation sequencing technology to sequence platelet RNA of healthy subjects and lung cancer patients,screen their differentially expressed genes,and verify them,laying a foundation for future application in lung cancer diagnosis.Methods: In the first part,peripheral venous blood 6ml was collected from 5 healthy subjects and 7 patients with lung cancer,from which platelets were isolated,and total platelet RNA was extracted by Trizol method.The total platelet RNA was sequenced using the next generation sequencing technology,and the gene expression was quantified by combining with the algorithm,and the differentially expressed genes were screened.Cluster analysis,Gene Ontology functional analysis and KEGG pathway enrichment analysis were performed on the differentially expressed genes.Six significantly differentially expressed genes were then selected as candidate genes.In the second part,4 healthy subjects and 6 lung cancer patients were included as validation subjects.Then the candidate genes were verified by real-time fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR).Results: Platelet RNA sequencing showed that there were 1076 differentially expressed genes in the experimental group compared with the control group.Functional analysis of Gene Ontology suggested that the differentially expressed genes were mainly related to vesicles,growth factor binding and other processes.The enrichment analysis of KEGG pathway showed that the up-regulated differentially expressed genes were mainly related to adhesion plaque,transcription disorders in cancer,and the interaction between cells and extracellular matrix and its receptors.The down-regulated differentially expressed genes were mainly related to programmed necrosis,tumor necrosis factor signaling pathway,and the interaction between cytokines and their receptors.Six significantly differentially expressed genes were selected,namely MAOB,MAP2,STK19,NR2F6,VAT1 and F8A3.The results of qRT-PCR were consistent with those of sequencing.Conclusions:1.This study sequenced platelet RNA from healthy subjects and lung cancer patients based on the next generation sequencing technology,and the results showed that there were significant differences in the gene expression profiles of platelet RNA between healthy subjects and lung cancer patients.2.qRT-PCR results showed that the expressions of MAOB,MAP2,NR2F6 and VAT1 in the lung cancer group were up-regulated compared with the healthysubjects,and the expressions of STK19 and F8A3 in the lung cancer group were down-regulated compared with the normal group.The results were consistent with the sequencing results,suggesting that these differentially expressed genes may be involved in the occurrence and development of lung cancer.
