Objective:To explore the expression of two possible hsa-mi R-135b-5p target genes,zinc finger ring protein 3(ZnRF3)and 6B(lysineacetyltransferase6B(KAT6B),in different gastric tissues and their correlation with the clinicopathological features of gastric adenocarcinoma,and to analyze the correlation between the expression of ZnRF3 and KAT6 B in gastric adenocarcinoma,so as to provide more possible clues for the diagnosis and treatment of gastric adenocarcinoma.Methods:Immunohistochemistry(IHC)was used to detect the expression of ZnRF3 and KAT6 B in 70 cases of gastric adenocarcinoma,31 cases of gastric cancer adjacent tissues and 30 cases of gastric precancerous lesions.The clinical and pathological data of gastric adenocarcinoma patients including tumor size,vascular invasion and nerve invasion were collected,and the data were collated and statistically analyzed by SPSS18.0.Results:(1)ZnRF3 protein was localized in cytoplasm.The positive expression rates of ZnRF3 protein in paracancerous tissues,precancerous lesions and gastric adenocarcinoma tissues were80.65%(25/31),70.00%(21/30)and 41.43%(29/70),respectively,with statistical difference(P=0.001).The positive rate of ZnRF3 in paracancerous tissues and precancerous lesions was higher than that in gastric cancer tissues(P=0.001/P=0.009),but there was no significant difference between paracancerous tissues and precancerous lesions(P > 0.05).The analysis of ZnRF3 protein and clinicopathological characteristics of gastric adenocarcinoma showed that there was a correlation between the expression level of ZnRF3 protein and TNM stage of gastric adenocarcinoma.the positive expression rate of ZnRF3 in stage I and II was 54.05%(20/37),which was higher than that in III and IV stages of gastric adenocarcinoma(27.27%).There was no significant difference between the expression of ZnRF3 protein and the pathological features of gastric adenocarcinoma patients including sex,age,tumor size,degree of differentiation,lymph node metastasis,vascular invasion and nerve invasion(P > 0.05).(2)KAT6B protein was localized in cytoplasm.The positive expression rates of KAT6 B protein in paracancerous tissues,precancerous lesions and gastric adenocarcinoma tissues were 67.74%(21/31),60.00%(18/30)and 32.86%(23/70),respectively,with statistical significance(P=0.002).The expression of KAT6 B in gastric adenocarcinoma was significantly lower than that in paracancerous tissues and precancerous lesions(P=0.001/P=0.011),but there was no statistical significance between paracancerous tissues and precancerous lesions(P=0.529).The analysis of KAT6 B and clinicopathological features of gastric adenocarcinoma showed that there was no significant correlation between the sex,age of onset,tumor size,TNM stage,lymph node metastasis,degree of differentiation,vascular invasion and nerve invasion(P > 0.05).(3)Spearman correlation analysis showed that there was no correlation between the expression of ZnRF3 and KAT6 B in gastric adenocarcinoma(P > 0.05).Conclusions:(1)The expression level of ZnRF3 and KAT6 B in gastric adenocarcinoma are lower than that in paracancerous tissues and precancerous lesions,but there is no significant correlation between the two proteins in gastric adenocarcinoma.(2)the expression level of ZnRF3 is related to the TNM stage of gastric adenocarcinoma,and the later the stage,the lower the positive expression rate of ZnRF3.
