| Objective: The research group intervened in SHR by Huanglian Jiedu Decoction,and detected the expression of mir-133 a,caveolin-1,e NOS m RNA,caveolin-1 and p-e NOS protein in aorta by q RT-PCR,WB,etc.,and intervened BAECs with HLJDD drug-containing serum.To verify the role of mi R-133a/Caveolin-1/e NOS pathway in "HLJDD up-regulation of SHR aortic e NOS/NO signaling",and provide experimental support for the clinical application of Qingrejiedu classic ancient prescription.METHODS: Twenty-five-week healthy male SPF SHRs weighing 150-200 g were randomly divided into 5 groups: 10 in the model group;10 in the high,medium and low dose groups in the HLJDD group;captopril Group 10;10WKY rats of the same age were blank control group.Coptis,Phellodendron,Astragalus,and Scorpion are composed of 3:2:2:3 ratio.The medicinal materials are weighed according to the prescription,and concentrated into extracts according to the method reported by Wu Hongyuan,etc.,and the feeding is started at different doses after one week of adaptive feeding.The course of treatment was 42 days,fasting for 12 hours,specimens were collected after anesthesia,and indicators were observed.Cell experiment:High,medium and low dose groups,captopril drug-containing serum group,blank control drug-containing serum group and normal group drug-containing serum were prepared.Bovine aortic endothelial cells(BAECs)were cultured with different concentrations of Huanglian Jiedu Decoction containing serum,captopril serum group with captopril containing serum,blank control serum group with blank control serum culture,normal group with routine methods,each group were cultured for 24 hours,observation indicators.Results:1 it has been proved that huanglian jiedu decoction can reduce SHR blood pressure,and the optimal concentration is high dose concentration.2 huanglian jiedu decoction can increase the expression of mir-133 a in SHR body and down-regulate the expression of proteins including Caveolin-1 and inflammatory factors of et-1.3.The contents of et-1 in cells in the captopril group and the huanglian jiedu decoction group and the high-dose group were significantly decreased,but the contents of NO and e NOS were increased.4.Mir-133 a and e NOS m RNA expression levels of cells in the high-dose huanglian jiedu decoction group were significantly higher than those in the blank serum group and the low-dose group.The expression levels of Caveolin-1 m RNA and Caveolin-1 protein in captopril group,huanglian detoxification decoction and high dose group were lower than those of blank control group.Conclusion:Huanglian jiedu decoction may over-express mir-133 a of SHR aorta,and regulate e NOS/NO signal through Caveolin-1,that is,the mir-133 a of huanglian jiedu decoction with high expression of aorta down-regulated Caveolin-1,and down-regulated Caveolin-1 further increased e NOS content and activity,and eventually led to up-regulation of NO signal. |
