| Objective: To observe the effect of Wenyang Zhuyu Decoction on the expression of stromal cell-derived factor-1(SDF-1)and its receptor CXC chemokine receptor 4(CXCR4)in brain tissue of rats with kidney-yang deficiency type cerebral ischemic injury.To explore the possible mechanism of Wenyang Zhuyu Decoction in preventing and treating cerebral ischemia injury.Methods: 192 healthy male SD rats were first divided into a blank group(n=48)and a kidney-yang deficiency model group(n=144).In the kidney-yang deficiency model group,the kidney-yang deficiency model was established by intramuscular injection of hydrocortisone injection for21 days,and the serum cyclic adenosine monophosphate(cAMP)was detected by enzyme-linked immunoassay(ELISA),the content of cyclic guanosine phosphate(cGMP).After successful modeling,144 rats with kidney-yang deficiency were divided into three groups: sham operation group,model group,and Wenyang Zhuyu decoction group,with 48 rats in each group.The blank group was fed normally and was given an equal volume of distilled water;the sham operation group was given only a0.8cm thread plug and given an equal volume of distilled water;the model group was given MCAO with an equal volume of distilled water;the Wenyang Zhuyu decoction group was given MCAO with an intragastric administration Wenyang Zhuyu Decoction.Samples were taken at four time points 3d,7d,14 d,21d after the start of gavage,and the content of cAMP and cGMP in serum were detected by ELISA;Western blot and Real-Time PCR were used to detect The mRNA and protein expression levels of SDF-1 and CXCR4 in brain tissue at each time point;HE staining method was used to detect the morphological changes of nerve cells on the ischemic injury side at each time point.Use SPSS24.0statistical software for data analysis.Results:(1)The changes of specific indexes of kidney yang deficiency:(1)after the model was completed,compared with the blank group,the serum cAMP level,cGMP level and camp/cgmp ratio of the rats in the model group were significantly lower than those in the blank group(P<0.01);(2)After 21 days of gastric administration,compared with the model group,cGMP level was significantly decreased in Wenyang Zhuyu Decoction group,cAMP level and camp/cgmp ratio were significantly increased(P<0.01).(2)The score of nerve function defect:(1)compared with the sham operation group,the nerve function defects of model group and Wenyang Zhuyu Decoction group were significantly different(P<0.01);(2)Compared with the model group at the same time point,the nerve function defects of the rats in Wenyang Zhuyu Decoction group were improved to different degrees(P<0.01 or P<0.05).(3)The morphological changes of brain tissue:(1)blank group and sham operation group: the cells were arranged in a regular manner,the membrane was intact,and the morphology and structure were basically normal;(2)Model group: the cell arrangement was disordered and sparse,and a large number of cells were necrotic,showing obvious ischemic damage;(3)Wenyang Zhuyu Decoction group: with the prolongation of the time of administration of stomach,compared with the model group,the cells increased significantly,the membrane was relatively complete,the cytoplasm was light purple,and the nucleus was gradually clear.(4)At the same time,the expression of SDF-1 and CXCR4 protein was compared with that of the blank group and sham operation group(P<0.01);(2)The expression of Wenyang Zhuyu Decoction group was higher than that of model group,the difference was statistically significant(P<0.01).(5)At the same time,the expression of SDF-1 and CXCR4 mRNA was compared with that of sham operation group(P<0.01);(2)The expression of Wenyang Zhuyu Decoction group was higher than that of model group,and the difference was statistically significant(P<0.01 or P<0.05).Conclusion: Wenyang Zhuyu Decoction significantly reduces the neurological deficit scores in rats with kidney-yang deficiency type cerebral ischemic injury.The mechanism of action may be to improve the kidney-yang deficiency syndrome on the one hand,and on the other hand to improve the cerebral ischemic state of the blood stasis cerebral artery,and then up-regulate the mRNA and protein expression levels of SDF-1and CXCR4 in the ischemic brain tissue,And then exert the protective effect of ischemic brain injury. |
