| Objective: The expression of miRNA-147a in ovarian cancer was detected to further explore the clinical significance of miRNA-147a expression in ovarian cancer.Methods: A total of 40 cases of ovarian cancer tissue and 40 cases of normal ovarian tissue were collected from the gynecology department of the First Affiliated Hospital of Wan Nan Medical University from October 2019 to October 2020.Fasting plasma samples were collected from all patients(40 ovarian cancer patients and 40 non-ovarian tumor patients)in the morning before surgery.Human EOC cell lines HO8910,A2780,SKOV-3 and human normal ovarian epithelial cell lines IOSE80 were purchased and frozen in a refrigerator at-80℃ for subsequent experiments.Real-time fluorescent quantitative PCR(q RT-PCR)was used to detect the expression and differences of miRNA-147a in histological specimens,plasma specimens and cells,and to understand the correlation between miRNA-147a expression and its clinicopathological characteristics in ovarian cancer.Results:1.The expression level of miRNA-147a in normal ovarian tissue was set as 1,and the relative expression level of miRNA-147a in ovarian cancer tissue was0.34±0.17,and the expression level of miRNA-147a in ovarian cancer was significantly decreased,and the difference was statistically significant(P < 0.05).2.When the relative expression level of miRNA-147a in tissues was higher than 0.34,it was considered high expression,and lower than 0.34 was considered low expression,it could be seen that the high and low expression level of miRNA-147a was not correlated with the age of ovarian cancer patients(P>0.05),but tumor pathological grading,FIGO staging and lymph node metastasis of correlation,the difference was statistically significant(P<0.05).3.The expression level of miRNA-147a in normal ovarian cell line IOSE80 was set as 1,and the relative expression level of miRNA-147a in human EOC cell line A2780,EOC cell line HO8910 and EOC cell line SKOV-3 were0.42±0.12,0.23±0.18 and 0.33±0.14,respectively,with statistical significance(P<0.05).4.Mi RNA-147a was expressed in the plasma of both groups of patients.When the preoperative plasma miRNA-147a expression level of non-ovarian tumor patients was set as 1,the relative preoperative plasma miRNA-147a expression level of ovarian cancer patients was 0.35±0.21,and the difference between the two groups was statistically significant(P<0.05).Conclusion:1.The low expression of miRNA-147a in epithelial ovarian cancer suggests that miRNA-147a may be a tumor suppressor gene.2.The expression level of miRNA-147a in epithelial ovarian cancer was negatively correlated with tumor pathological grading,FIGO stage and lymph node metastasis,suggesting that miRNA-147a may play an inhibitory role in the progression of epithelial ovarian cancer. |
