Objective: The bioinformatics analysis method was used to mine and analyze the colon cancer data from The Cancer Genome Atlas(TCGA),and six immune-related lncRNAs related to prognosis were screened.The prognostic value of the model constructed by immune-related lncRNAs was evaluated by survival analysis and univariate and multivariate Cox analysis.Methods: The RNA data of colon cancer patients were obtained through the TCGA data website.Combined with the human reference genome,the gene data with gene name and sample name were obtained,and the expression data of m RNA and lncRNA were obtained respectively.Then,the immune-related genes were obtained from the molecular signature database,and the immune-related lncRNA of colon cancer patients was obtained through the co-expression network.Through the single-factor Cox and multi-factor Cox regression analysis of lncRNAs,six lncRNAs are finally determined to construct the risk model.According to the risk score,the patients were divided into high-risk group and low-risk group.The survival difference between the two groups was verified by survival analysis,and the prognostic value of the model was evaluated by comparing with other clinical factors.Results: 1.19 immune-related lncRNAs related to prognosis could be obtained by univariate Cox regression analysis.2.Through multivariate Cox regression analysis,six lncRNAs were established to construct the risk model,in which SNHG16 and LINC00513 were considered as protective effect factors of HR <1,and the other four lncRNAs : AL354993.2,AC138207.5,PRR7-AS1 and AC090517.2 were considered as risk effect factors of HR >1(p < 0.05).3.Patients with colon cancer can be divided into high and low risk groups according to the score calculated by the risk model,and there is a statistically significant difference in the survival status between the two groups(p < 0.001).Conclusion: 1.According to the survival data and RNA expression profile of TCGA patients,combined with immune gene bank,immune-related lncRNAs,related to the prognosis of colon cancer can be obtained.Based on this,the colon cancer risk model is constructed and the risk score formula is obtained.2..This lncRNAs model can help predict the prognosis of patients with colon cancer. |
