Objective:Based on the public database of TCGA(The Cancer Genome Atlas)of high throughput datam-RNA expression profile chip,this study explored the changes of gene expression level induced by Colon Cancer(CRC),did coexpression network analysis of differential expression genes and estimated the survival period of colon cancer patients,so as to provide scientific basis for exploring the molecular mechanism and prevention analysis of CRC.Methods:The colorectal cancer gene expression profiles datasets were downloaded from the public reading TCGA website(http: //cancer genome.nih.gov/),including 32 cases of colon cancer(colon cancer)and 32 cases of adjacent tissues(adjacent group)of the m-RNA sequencing data level 3 raw counts and RNA-Seq by Expectation Maximization RSEM normalized read counts and the m-RNA and clinical information of 507 patients with colon cancer.After the merger,integration and quality control of the 20531 genes in 32 paired data,I used R software to program m-RNA differently expressed genes analysis,heat maps,coexpression network analysis of differential expression genes and survival analysis of 507 cases of colon cancer patients.Results:2939 genes with significant difference in the expression of in colorectal cancer were screened from 20531 genes in 32 cancer and adjacent groups using the Bayesian method and the multiple analysis method in the colon cancer data set using the limma package.Coexpression network analysis of 2939 differential expression genes(1533 up-regulated genes and 1406 down-regulated genes)finally found 10 gene modules.507 Colon Cancer patients were estimated to have a median survival period of 7.57 years and95% confidence intervals(3.93,9.62)years.Conclusion:In this study,the differently expressed genes of colon cancer and coexpression modules were found using the public database TCGA for the further research of colon cancer genome. |
