| Objective To explore the relationship between Serum 25 hydroxyvitamin D3(25-OH-D3)level and premature coronary heart disease(PCHD).Methods55 patients with restenosis after coronary stent implantation in the First Affiliated Hospital of Jinzhou Medical University from 2009 to 2019 were selected as the study group,and 70 patients without restenosis after stent implantation were selected as the control group.Pittsburgh sleep quality index(PSQI)was used,PSQI was used to evaluate sleep quality.The metabolites in the blood of the two groups before coronary angiography were detected by tandem mass spectrometry.Metabonomics analysis was performed between the two groups.Principal component analysis(PCI)and orthogonal partial least squares discriminant analysis(opls-da)were used to analyze and compare the differences of metabonomics between the two groups.The serum levels of IFN-γ,IL-2,IL-4 and IL-10 were measured.Statistical analysis(spss25.0,SPSS,USA software and simca-p14.1,Sweden,umetrics).The mean ± standard deviation(x ± s)was used to represent the measurement data.Principal component analysis(PCA)and orthogonal partial least squares analysis(opls-da)were constructed after pretreatment of metabolomic data between groups,and permutation was used to verify the validity of the model.The differential metabolites were screened according to VIP value and VIP confidence interval,and then the screened variables were tested by independent sample nonparametric test.Results The results of Pittsburgh sleep quality index(PSQI)in the restenosis group showed that the sleep quality,sleep time,sleep efficiency,time function and total score of PSQI were higher than those in the control group;there was no difference in sleep disorder and hypnotic drugs between the two groups.PCA was used to analyze the plasma metabolites of the two groups.The results showed that the metabolites had a relatively obvious trend of differentiation and clustering(r2x = 49.0%,Q2 = 11.8%).A total of 16 metabolites were screened and identified in opls-da model,of which 4 were related to amino acid metabolism and 12 were related to carnitine metabolism.The levels of IFN-γand IL-2 in the restenosis group were higher than those in the control group,while the levels of IL-4 and IL-10 in the restenosis group were lower than those in the control group Correlation analysis showed that the total score of PSQI was positively correlated with the levels of IFN-γ and IL-2 in restenosis group.Conclusions1.The relationship between abnormal sleep and stent restenosis is pure in a certain degree.2.One month after operation,the metabolites could distinguish the STEMI patients in group A and group B.3.In group B,the contents of leucine,valine(branched chain amino acids,BCAA)and various acyl carnitines were relatively high,and the cardiac function was relatively poor.It is speculated that the above metabolites may participate in the process of cardiac dysfunction after myocardial infarction,promoting BCAA catabolism and carnitine treatment can improve the process of heart failure after myocardial infarction. |
