| Object:Through the preliminary investigation of P.cuspidatum resources in Hubei Province,this study understood the distribution of wild resources of P.cuspidatum in Hubei Province,investigated the planting area and yield of cultivated P.cuspidatum in Hubei Province,and understood the market demand of the province,so as to provide reference for the rational and orderly development of P.cuspidatum industry.At the same time,the quality of P.cuspidatum in Hubei Province was evaluated and high-quality resources were selected through routine inspection,HPLC fingerprint establishment,and content determination of main active components.In addition,this study investigated the correlation between the content of principal components and color,and established a quantitative model for the rapid and accurate determination of water content and extract for the rapid detection of related indexes of the medicinal materials,aiming to provide a new method for the quality evaluation of P.cuspidatum.Methods:1.Preliminary investigation of wild resources and cultivation status of P.cuspidatum in Hubei Province through literature review,investigation and field investigation.2.The quality of P.cuspidatum was evaluated by measuring the content of water,ash,acid-insoluble ash and alcohol-soluble extract.3.By establishing HPLC fingerprint of P.cuspidatum,cluster analysis,principal component analysis and partial least square analysis were used to evaluate the comprehensive quality of P.cuspidatum.4.The content of heavy metals in P.cuspidatum and its growing soil were determined by ICP-MS and atomic absorption spectrometry.Correlation analysis was used to evaluate the environmental quality of heavy metals in soil and the absorption characteristics of heavy metals in P.cuspidatum.5.The calibration set samples were scanned,analyzed and modeled by Fourier transform NIR spectrometer combined with partial least squares(PLS)to establish a quantitative model for the determination of moisture and extract.6.The color difference value of P.cuspidatum powder was measured by color difference instrument,and the correlation analysis was conducted with the content of main components of P.cuspidatum.Results:1.The wild resources of P.cuspidatum were distributed in all regions of Hubei province,with scattered producing areas,mainly distributed in Shiyan,Huanggang,Enshi,Yichang and other places,with the altitude of 5~1860 meters.At present,the cultivation area of Polygonum kepticum in Hubei Province is up to 47,800 mu,Now every three years is a harvest period,which can produce 57400 tons of dry goods.mainly concentrated in Fangxian County,Huanggang City,Enshi City and other places.Among them,Shiyan Fangxian County has the largest cultivation area of 20,700 mu.Two giant knotweed processing enterprises in Hubei Province need 20,000 tons of dried P.cuspidatum annually,and the demand is relatively stable and in a state of balance between supply and demand.2.The water content and alcohol-soluble extract content of 78 batches of P.cuspidatum reached the pharmacopoeia standard.The total ash content of 16 batches of medicinal materials exceeded the limit value,and the acid insoluble ash content of 7 batches exceeded the limit value.3.Nine common peaks were identified by HPLC fingerprint of P.cuspidatum,and five compounds were identified by comparison with reference materials.The similarity ranged from 0.628to 0.996,indicating that different growth years,harvesting time and producing areas resulted in great differences in components of P.cuspidatum.Content determination combined with routine inspection showed that the quality of P.cuspidatum in Songzi area of Jingzhou was superior.The results of principal component analysis and partial least square analysis showed that emodin-8-β-O-D glucoside and resveratrol were different compounds between wild and cultivated P.cuspidatum.4.The results of heavy metal content in 36 batches of P.cuspidatum soil and medicinal materials showed that the content of Cd in samples from Xuejiaban Village,Yingshan Nanhe Town,Huanggang,and Gujia Agricultural P.cuspidatum from Yingshan,Huanggang was more than0.3mg·kg-1,while the content of heavy metals in other areas was low.The results of statistical analysis showed that P.cuspidatum had a strong ability to enrich Cd in the growing soil,which should be paid more attention to in the production.5.The model of rapid determination of water and alcohol soluble extract in P.cuspidatum by NIRS was established by internal cross test.The R~2 of internal cross validation for water and extract were 90.88 and 96.15,respectively.External validation of the prediction set showed that the correlation coefficients R were0.9910 and 0.9847,respectively.6.The three anthraquinones in P.cuspidatum were significantly correlated with the color index L*(representing color brightness)and E*ab(total color difference)(P<0.01),indicating that,to some extent,the lower the value of L*and E*ab,the higher the contents of the three anthraquinones.Conclusion:The similarity of Huanggang Qichun(YS-31)was only0.628,when the genetic distance was large,it could always be grouped into one kind alone.The similarity of 1-year-old P.cuspidatum fingerprint was 0.671.The clustering results showed that most of the cultivated and wild P.cuspidatum in Hubei Province were clustered in the same group,which indicated that there was no significant difference between the wild and the wild,but the partial least square method showed significant difference in the contents of emodin-8-β-O-D glucoside and resveratrol,further verification is required later.The results of content determination and inspection showed that the quality of P.cuspidatum produced in Songzi of Jingzhou,Jundian of Fangxian and Longtanfan of Yingshan was superior.We should strengthen the monitoring of Cd content in soil and the basic inspection of P.cuspidatum.It is feasible to determine the content of water and alcohol soluble extract in P.cuspidatum by using the model of near infrared spectroscopy combined with chemometry,which provides a new idea for rapid and non-destructive quality control of P.cuspidatum and its preparation.The content of three anthraquinones in P.cuspidatum powder can be predicted by measuring the color difference value of P.cuspidatum powder with color difference meter. |
