As a commonly used traditional Chinese medicine,Polygonum cuspidatum has a long history of medication.Ancient books record many processing methods of Polygonum cuspidatum,from Shennong materia classic of the Eastern Han Dynasty to compendium of Materia Medica of the Ming Dynasty,which are mainly based on "fine file,sun dry",and there are also different processing methods such as "wash off the chapped skin,file,bake and screen","burn,file",but the specific processing technology is not described in detail.At present,the processing method of Polygonum cuspidatum in Chinese Pharmacopoeia 2015 is "slice thick,dry",and there is no specific description of the process;literature research shows that Polygonum cuspidatum cuspidatum has a good clinical effect on gout,but its material basis of efficacy is not clear.Therefore,this paper studies the processing technology and the material basis of anti gouty arthritis of Polygonum cuspidatum,in order to standardize the processing technology of Polygonum cuspidatum and clarify the main active components and mechanism of anti gouty arthritis of Polygonum cuspidatum.The main research contents are as follows:1.Optimization of processing technology of Polygonum cuspidatum by single factor experiment and orthogonal experimentFirstly,the medicinal materials of Polygonum cuspidatum were divided into two grades.Then,the single factor experimental design method was used.The soaking time,moistening temperature and moistening time were used as the factors.The extract,ash,Polygonum cuspidatum glycoside,resveratrol,rhubarb 8-o-?-D glucoside,emodin and emodin methyl ether were used as the evaluation indexes.The differences among the five effective components of each group were analyzed by cluster analysis Combined with the data of leachables and ash,the best softening process was selected.The drying process of Polygonum cuspidatum was designed by orthogonal experiment.The factors of drying time,drying temperature,drying thickness and turning times wereinvestigated.The evaluation indexes were water,extract,Polygonum cuspidatum glycoside,resveratrol,rhubarb 8-o-?-Dglucoside,emodin and emodin methyl ether.The weight coefficients of each index were calculated by AHP software,and then the comprehensive weighting was used Excellent scoring method.The drying technology of Polygonum cuspidatum was selected,and the softening and drying technology of Polygonum cuspidatum was verified.The best processing technology of small-scale Polygonum cuspidatum is: soak it for 30 minutes,moisten it at20 ? for 24 hours,cut it into thick slices,dry it at 60 ?for 4 hours,dry the thickness of 3cm,and turn it once during drying;the best processing technology of large-scale Polygonum cuspidatum is: moisten it at 20 ? for 45 hours,cut it into thick slices,dry it at 60 ? for 4 hours,dry the thickness of 4cm,and turn it once during drying.2.Study on the relationship between HPLC fingerprint of Polygonum cuspidatum and spectrum effect of anti gouty arthritisThe HPLC fingerprint of Polygonum cuspidatum was established by analyzing 10 batches of Polygonum cuspidatum pieces.The thickness of the foot pad and the UA content in theserum of mice were taken as the effective indexes of Polygonum cuspidatum against gouty arthritis.The relationship between the spectrum and the effect was analyzed by the method of grey correlation analysis.It was found that the anti gouty arthritis effect of Polygonum cuspidatum might be the result of multi-component interaction.The main characteristic peaks of anti gouty arthritis were peak 7(polydatin),peak 9(quercetin),peak 11(resveratrol),peak 12(quercetin),peak13(emodin-8-o-?-D-glucoside),peak 16(emodin)and peak 18(emodin methyl ether).3.Pharmacodynamic study of Polygonum cuspidatum glycoside on gouty arthritisSD rats were randomly divided into six groups: blank control group,model group,low-dose group,middle dose group,high-dose group and colchicine group.Except for the blank control group,other groups were subcutaneously injected with0.2 ml(4 mg)of endotoxin free MSU crystal suspension into the right foot pad to induce inflammation.The rats in the low,middle and high dose groups were given 25,50 and 100 mg / kg by gavage,while those in the colchicine group were given 1 mg/ kg by gavage.The rats in the blank control group and the model group were given the same volume of normal saline by gavage.Continuous administration for 3 days.To evaluate the effect of polydatin on gouty arthritis,the swelling degree of foot pad thickness,the content of serum IL-1 ?,IL-6,PGE2,MCP-1,UA,MPO,no,XOD were studied.Compared with the model group,each administration group can significantly reduce the swelling degree of SD rat's foot pad,and the low,medium and high dose groups of polydatin can reduce the content of IL-6,IL-1 ?,UA,XOD,no,PGE2,MPO and MCP-1 in varying degrees,among which the high dose group has the best effect(P < 0.05).These results suggest that polydatin may play an anti gouty role in three aspects: purine metabolism,uric acid metabolism and inhibition of inflammatory cytokines.
